One of the most applied methods in drug industry for development of new drugs is 3D-QSAR methodology. As p38-mitogen-activated protein kinase (p38-MAPK) plays a crucial role in regulating the production of such proinflammatory cytokines as tumor necrosis factor-α (TNF-α) and interleukin-1, emerging as an attractive target for new anti-inflammatory agents, we used a 3D-QSAR based method of Comparative Molecular Field Analysis (CoMFA) on a series of 52 potent p38-MAP kinase inhibitors with IC50 ranging from 3.2 to 10,000 nM. An alignment rule for the compounds was defined using Distill in SYBYL 7.3. The best model was validated using a data set obtained by dividing the data set into a training set and test set using hierarchical clustering, based on the CoMFA fields and biological activities (pIC50). The best predictions were obtained with a CoMFA region-focusing model (R2 ncv = 0.952, q2 = 0.678, R2 Pred = 0.627). The statistical parameters from the model indicate that the data are well fitted and has high predictive ability. Moreover, the resulting 3D CoMFA contour maps provide useful guidance for designing highly active inhibitors. پرونده مقاله

Ibuprofen (I) is the most widely used non-steroidal anti-inflammatory drugs (NSAIDs) has been synthesized in recent years. NSAIDs are the first choice of drugs that are normally used for the treatment of pain and inflammation. There are many methods for synthesizing of I but each one has some difficulties as raw materials preparation, reaction conditions and etc. In an effort to establish modified method with improved and economical yield, I was synthesized by the mesylation of ethyl lactate in the presence of triethylamine or pyridine at 273 K to obtain the corresponding ethyl-2-(methylsulphonyloxy) propanoate (II). Then, Friedel-Crafts alkylation of II with isobutylbenzene for single step synthesis of ethyl-2-(4- isobutylphenyl) propanoate (III) was carried out by heating with AlCl3 under neat reaction conditions. Finally, ethyl-2-(4-isobutylphenyl) propanoate (I) formed in 50 % yield was hydrolyzed with KOH in methanol and acidified with HCl to afford the desired product. پرونده مقاله

Alpinia galanga and Alpinia calcarata are two important aromatic medicinal plants belong to the family Zingiberaceae. In the present study, dried rhizomes of A. galanga and A. calcarata were extracted with water and then fractionated with different solvents. Among the various solvent fractions, ethyl acetate (EA) fraction of both the plants showed high polyphenolic content with better antioxidant activity. This polyphenol rich EA fraction is also screened for its antidiabetic and anti-inflammatory potential using in vitro assays. Results showed that EA fractions of these two plants had significant antihyperglycemic activity by inhibiting carbohydrate digesting enzymes (α-amylase and α-glucosidase) and by inhibiting glycation reaction. Inhibitory potential of EA fractions of both the plant against protein denaturation and xanthine oxidase validates its anti-inflammatory potential. Moreover, EA fractions also exhibited significant LDL oxidation inhibition .The observed antidiabetic and anti-inflammatory properties of A. galanga and A. calcarata could be due to the synergic effect of polyphenols such as gallic acid and ellagic acid as well as bioactive alkaloid, berberine. پرونده مقاله

Pterospermum rubiginosum B.Heyne ex G.Don (PR) is a traditional medicinal plant used by the tribal people of the Western Ghats to treat bone fractures, inflammation, and sprain. Being an under-explored medicinal plant, the mechanism behind the anti-inflammatory activity of PR is unknown to the scientific community. FTIR analysis was done to recognize the functional groups in PR bark extract, and LCMS elucidated the phytochemical characterization. Molecular docking studies showed an excellent ligand and protein (inflammatory mediators) binding and their interactive mechanism. Total phenolic and total flavonoid contents were found to be 46.7 and 37.4 g/100 g for PRME. PRME showed hydrogen peroxide and DPPH (1,1diphenyl-2-picryl hydrazyl) radical scavenging activity with IC50 values of 48.33 and 42.70 µg/mL, respectively. Gene expression study of COX-1 and COX-2 enzymes confirmed the anti-inflammatory activity of PRME. The present study justifies the potential use of PR in the traditional medicine as an anti-inflammatory agent. پرونده مقاله

The tree Cryptocarya concinna Hance is largely distributed in southeast Asia and essentially used for its robust wood. Alcoholic extracts made from the leaves or roots of the plant have revealed anti-inflammatory and anticancer properties. The DNA-damaging dihydrochalcone derivative cryptocaryone accounts for the anti-proliferative and pro-apoptotic activities observed with the extracts. Other bioactive products have been isolated, including α-pyrone derivatives such as cryptoconcatones A-L and flavonoids such as cryptoconones A-E. A structural analogy is underlined between cryptoconcatone D and two known compounds with an α,β-unsaturated δ-lactone: pironetin and the styryl-lactone goniothalamin which targets the peroxisomal protein MFE2 (multifunctional enzyme type2). Pironetin is a potent α-tubulin binder with robust anticancer properties. By analogy, the binding of cryptoconcatone D to the pironetin-site of α-tubulin is proposed. The review shed light on the phytochemical constituents of Cryptocarya concinna and the biological properties of α,β-unsaturated lactone compounds isolated from this little studied plant. پرونده مقاله

Essential oils (EOs) are concentrated liquids extracted from various parts of plants and can be classified based on their phytochemical compounds. Terpenes and terpenoids have a wide range of biological activities, including anticancer, anti-inflammatory, antimicrobial, antioxidant, and antiallergic properties. Terpenes are plant-based compounds commonly used in the pharmaceutical, food, biofuel, and chemical industries by humans. In synthetic biology, genomic resources and emerging tools facilitate the production of high-quality terpenoids in plants and microbes. Terpenoids, however, are difficult to produce in large quantities due to their complex chemical structures and the limited amounts found in plants. The regulation of terpenoid biosynthesis has gradually emerged as a research priority. This review presents an overview of the biological activities, synthesis pathways, and key enzymes involved in the biosynthetic pathways and regulation of terpenes or terpenoids. This review will also include references for further research on molecular regulation, biological advancements, and increasing the content of terpenes or terpenoids in plants. پرونده مقاله

Background & Aim:Zea mays leaf used traditionally in Ibibio ethno medicine in Southern Nigeria to treat malaria and other inflammatory diseases. These anti-inflammatory and analgesic properties were evaluated against experimentally induced inflammation and pains using standard models. Experimental: The effects of ethanol leaf extract of Zea mays (170 -510 mg/kg) on experimentally induced inflammation and pain were studied in rats using Carrageenin, egg albumin and xylene-induced inflammation as well as acetic acid-induced writhing and thermal induced pain models. The GCMS analysis of the most active fraction was carried out. Results: The extract (170 -520 mg/kg) inhibited inflammation and pains caused by different phlogistic agents used in a dose dependent fashion. The various degrees of inhibitions were statistically significant (p<0.05, 0.01, 0.001), though incomparable to that of the standard drugs used (ASA and indomethacin). The GC-MS revealed the present of polyunsaturated fatty acids and phenolic compounds which may be responsible for the observed activities of the extract. Recommended applications/industries: The present study demonstrates that leaf extract of Zea mays might to be useful for the treatment of inflammation and pains. پرونده مقاله

Background & Aim: The use of anti-nociceptive and anti-inflammatory drugs in clinical medicine is necessary and unavoidable. Since the use of synthetic drugs is associated with numerous side effects, the application of complementary therapies such as herbal medicine can accompany with less side effects while having anti-inflammatory and analgesic properties. Satureja bachtiarica contains different active compounds such as Thymol, Para syman, Menton and Carvacrol which may be involved in the control of nociception and/or inflammation. Experimental: 75 male mice (weighing 30 -20 g) were divided to 15 groups. Analgesic and anti-inflammatory effects of hydroalcoholic Satureja bachtiarica extract at doses of 400, 600, 800 mg/kg were evaluated by writhing, formalin and xylene tests. Morphine (10 mg /kg) and dexamethasone (10 mg/kg) were considered as positive controls in analgesic and anti-inflammatory tests, respectively. Results & Discussion: Different doses of hydroalcoholic extract of Satureja bachtiarica significantly reduced pain in comparison to control groups, however, the anti-inflammatory effect of this drug evaluated by xylene test, has not been established which needs further examination through other tests. Industrial / Practical recommendation: According to present results, the analgesic effect of Satureja bachtiarica was comparable to morphine. However, it seems that further research is needed in the future to take advantage of this plant in relieving pain. پرونده مقاله

A new series of transition metal complexes of Co(II), Ni(II), Cu(II) and Zn(II), has beensynthesized from the Knoevenagel condensate Schiff base ligand(L) derived from β-ketoanilideand furfural with o-phenylenediamine and diethylmalonate. Structural features were determinedby spectral and analytical techniques. Square-planar geometry has been adopted by thecomplexes except cobalt complex which has an octahedral geometry. The synthesized ligand andits complexes were screened for their anti-inflammatory activity in male albino rats(Carrageenan-induced rat paw oedema model). Among these complexes, copper complex showssignificant anti-inflammatory activity. It exhibits significant dose dependent activity in acuteinflammation. The doses of 100 and 200 mg kg-1 bw produced 38.3 % and 42.8 % inhibitionrespectively after 3 h as compared with that of the standard drug (indomethacin) which showed48.5 % inhibition. The in vitro antimicrobial activity of the ligand and its complexes wasmonitored by disc diffusion method. It has been found that complexes have higher antimicrobialactivity than that of free ligand. پرونده مقاله

Several studies have been conducted to investigate the effects of herbal drugs and Non-steroidal anti-inflammatory drugs on the primary dysmenorrhea, but their results have been different. The aim of this study was to compare the effects of herbal drugs and NSAIDon the primary dysmenorrheausing meta-analysis method.A literature search was conducted in Scopus, Pub med, and Google scholar databases from1996 to December 2016 in English and Persian languages using the search keywordsof primary dysmenorrhea combined with herbal and NSAID. The random effect model was adoptedto combine the results of the studies, and I2 index was used to check heterogeneity. One hundred and ninety-nine studies were obtained, and after reading the abstracts, 21 articles were included in the study. The total number of samples was 2041 people. The standard mean difference(SMD)betweenherbal drugs and Non-steroidal anti-inflammatory drugs in the pain reduction in the first intervention was estimated atSMD= 0.2(95%CI:(-0.340.74 ), z=0.73, p=0.463), and in second intervention it was SMD=-0.59(95%CI: (-1.650.47),Z=1.09 and p=0.278). also, the SMD for the duration of menstrual variable SMD=0.18(95%CI:( 0.02 0.34) , z=2.20, p=0.027) in favor of NSAID drugs and interval cycle of menstrual variable was SMD=0.07(95%CI:(-0.140.28), z=0.68, p=0.496).Mean difference of pain intensity reduced relatively better by administering herbal medicines, which is supported by the majority of studies, and the herbal drugs were found to be more effective than the NSAIDs drugs. Finally, given the many side effects of non-steroidal anti-inflammatory drugs, herbal drugs can be used at the standard dosageand considering the side effects. پرونده مقاله
