        1 - Retrospective survey of abundance nematodes in digestive tract of sheep in East Azerbaijan and calculation of correlation between EPG and humidity in mentioned regions

        Abstract Infections with gastrointestinal (GI) nematodes severely affect small ruminant health and compromise their productivity and reproductive performances and can be a major cause of economic losses in small ruminant production. High burden of infections with nematodes may lead to death; and under field conditions, most infections are usually mixed consisting of different species of nematodes. Sheep and goats are infected with gastrointestinal nematodes while grazing and the biology of the infective larvae on the pastures is important in planning effective control programs. In general, the development of the free-living stages of gastrointestinal nematodes is largely dependent on temperature, with development rate increasing at warmer temperatures, while moisture is also required for the development cycle to proceed. Temperature also influences grass growth and hence exposure independently of larval development and survival. East Azerbaijan province in terms of livestock animal population in particular sheep breeding is one of the most important provinces of Iran and due to the amount of rainfall is susceptible to worm’s contamination in livestock animal. In this survey, fecal samples of 5934 sheep in 19 cities in East Azerbaijan, from 2002-2009 were examined by the Clayton-Lane method. Briefly, a sample of feces, 1-5 g, was well mixed with water (about 30-50 ml) and strained through a sieve (1 mm mesh) to remove coarse fecal material. The mixture was sedimented for 10-15 minutes on the bench, or by light centrifugation for two or three occasions, until the supernatant was clear. The sediment was then mixed with a saturated solution of sugar in a centrifuge tube (15 ml volume) and centrifuged for one or two minutes at 500 g. The egg floated and coverslips were placed on the tubes in centrifuge and after 5 min the samples were examined for the presence of eggs at 100x and 400x magnifications under the light microscope and the number eggs of Nematodirus, Marshallagia, Trichuris, unknown nematodes parasite eggs and the whole of the nematode eggs were determined in these cities by their shape and size. These data were analyzed by the SAS software. The results showed that the Kaleibar was the most infested city and the Malekan had the lowest contamination to gastrointestinal nematode parasites. Marshallagia and Trichuris had the maximum and minimum quantities of means with 11.12±32.02 and 2.23±13.78, respectively. A study on the correlation between humidity in the second month and humidity average and fecal egg count showed that there is a positive and significant (p < 0.01) correlation between them. Base on the results of this survey high humidity in regions and seasons are risk factors for propagating worm infestations and distinct handlings are necessary for preventing the mentioned phenomenon. This study survey on geographical situation in 19 cities of East-Azerbaijan indicated that the infestation rate to gastrointestinal nematodes in south of province (Malkan city) is at the minimum rate and in north province (Kaleibar city) is in maximum rate. Because the north region of East-Azerbaijan province is warmer and moister from the other regions, the high rate of infestation to GI nematodes was expected. Furthermore, differences in the population of livestock animals in the two regions are important in the rate of infestation to nematodes. In conclusion, the results of the study denoted that even approve the anthelminthic drugs in the livestock animals, the rate of infestation to GI nematodes is high. Also, the results of this survey are showed that a positive correlation is between EPG and the mean of humidity and this is important that treatment and prophylaxis are needed to decrease the risk of the disease. This research serves as a baseline for future studies on prognosis and prophylaxis of GI nematodes. These regional differences appeared to be systematic and can be ascribed to differences in applied pasture management and climatic conditions, and cause a need for regionally adapted monitoring and control systems.         2 - Determination of infestation rate of Ixodidae ticks to Theileria annulata in Urmia region by polymerase chain reaction (PCR)

        Theileriosis is one of the most important protozoan parasitic diseases in Iran that causes economic losses and mortality in livestock. The disease is transmitted by Ixodid ticks and is characterized by fever, anemia, anoxia and eventually death. Diagnosis of the disease was previously based on staining of blood and lymph node samples by Giemsa method. The use of molecular methods is preferred over previous methods due to their high sensitivity and specificity. Due to the fact that Ixodid ticks that can carry the disease are found in many traditional Iranian farms, it is very important to identify and introduce these ticks. The aim of this study was molecular identification of Theileria annulata infection in ticks isolated from cows in Urmia region. A total of 100 ticks from herds with a history of Theileria annulata infection in the area were collected and transferred to the laboratory. Ticks were identified using diagnostic keys. Salivary glands of ticks were separated and analyzed by PCR after isolation using Theileria annulata specific primer (N516, N517). Out of 100 ticks, ten genera and species including Hyalomma anatolicum anatolicum (33%), Hy. detritum (16%), Boophylus anulatus (16%), Dermacentor marginatum (14%), Rhipicephalus bursa (9%), Hy. anantolicum excavatum (7%), D. niveus (4%) and Haemaphysalis punctate (1%) were identified. The predominant tick was identified as Hyalomma anatolicum anatolicum. PCR test showed the presence of 721 bp specific band on agarose gel in 62% of tick samples. The high rate of infection in vector mites in the area indicates their high potential for infection of cows with the agent of Theileria annulata. Manuscript profile

One of the most important animal species living in it is the wild goat (Capra aegagrus). The aim of this study was to determine the level of parasitic infections of the gastrointestinal tract goats of Arasbaran National Park. During 2018, 95 fecal samples were collected from the wild goats of Arasbaran National Park and were examined by direct smear and Clyton-Lane methods. Bearman apparatus was also used to identify the first stage larvae of pulmonary nematodes. Iodine and modified Nelson staining were used to diagnose Cryptosporidium and Giardia infections, respectively. All samples showed at least one worm agent and one protozoan infection. In this study, Eimeria (spp.) infection was observed in all fecal samples and infection with egg of Marshallagia marshalli (37.8%), Nematodirus (spp.) (60%), Trichioris ovis (62.1%), Dicrocoelium dendriticum (8/37%), Fasciola hepatica (25.2%), Moniezia expansa (13.6%), Cystocaulus oecratus larvae (28.4%) and Giardia lambelia cyst (33.6%) were observed. Cryptosporidium oocysts were not observed in any of the stained stool samples. No significant relationship was found between infestation rate with different parasites and the sex of the infected animals. The high level of infection of parasites in wild goats of Arasbaran National Park indicates the high potential of parasitic infection and conversion of animals in it as a reservoir of parasites and its subsequent spread to other animals and expansion in neighboring human communities. This disease was identified for the first time in 1984 in Norway. The dog is the only definitive host and also the intermediate host of protozoa and contamination can also be seen in cattle, sheep, goats, horses, and deer. The importance of neosporosis disease is related to causing abortion in ruminants (especially cows) and the economic losses caused in industrial livestock farming, which has caused many studies to be conducted in Iran and rest of the world. The disease was first reported in Iran in 2004, and since that year, many studies have been conducted about it. In this review article, an attempt has been made to introduce the protozoa that causes the disease and its evolution, as well as the methods of diagnosing the disease, and provide an overview of the studies conducted on this disease in Iran, from the beginning to the present day.         This survey was conducted to study the seroepidemiology of hydatidosis in northwest of Iran. Totally 852 blood samples from rural inhabitants of Sarab, Meshkin shahr, Moghan and Urmia were collected and their sera were separated. For antigen preparation, the livers and lungs of sheep infected with hydatid cyst and slaughtered at Tabriz abattoir were collected. The protoscolex was separated from the hydatid cyst and injected into the rabbit diaphragm using the sandwich technique. Then sections of the diaphragm were cut using the cryocut apparatus and fixed onto slides. The prepared sections were examined using marked antihuman globulin stained with Evan’s blue by IFA test. Initially sensitivity and specifity of this test was evaluated on sera of 58 hospitalized hydatidosis patients and 70 healthy persons. The sensitivity and specifity of the test were estimated 88% and 98/6% respectively. In this survey the titers over 1:10 were considered positive. Among 852 tested samples, 30(3/52%) were infected. Infection rate in males was higher than females. The highest infection rate was from Meshkin shahr (3.5%) and the age group of 20-40 years was the highest infected category. Manuscript profile
