Leptospirosis is a common zoonosis caused by Leptospira interrogans. Leptospira are more durable in temperate and humid climates and if transmitted to a vertebrate host, can lead to clinical to subclinical disease with a wide range of symptoms including fever, jaundice, hematuria, abortion and sometimes death. Sheep are important hosts of Leptospira that have received less attention due to the higher prevalence of subclinical form of the disease in this species. The aim of this study was to determine the prevalence of leptospirosis serology in sheep in Khuzestan province using standard microscopic agglutination test (MAT).261 blood samples were taken from sheep in 10 cities of Khuzestan province during the autumn and winter of 2022. All samples were tested by MAT method using five serotypes of Leptospira Grippotyphosa , Pomona, Icterohemorrhagiae, Canicula and Hardjo. The results of MAT showed that 12 samples (4.5%) had positive reaction, of which 4 samples with Grippotyphosa, 4 samples with Icterohemorrhagiae, 3 samples with Hardjo and 1 sample with Pomona. Titration of positive samples showed that 10 samples had a titer of 1:100 and 2 samples had a titer of 1:400.The results of this study indicate a decrease in the serological prevalence of leptospirosis in sheep in Khuzestan province compared to previous studies. It seems that the trend of climate change, the unprecedented decrease in rainfall expectations over the past years and the increase in compliance with health protocols, have played an effective role in reducing the prevalence of the disease. Leptospirosis is a multi-faceted global disease, which causes great economic and health losses to the livestock and human population of our country each year. The northern provinces of Iran are among the rainy and humid regions of Iran, and the prevalence of waterborne diseases such as leptospirosis is high in these regions. In ruminants, cattle are more susceptible to leptospirosis than small ruminants. The purpose of this study was to determine the serological prevalence of leptospirosis in slaughtered cows in Guilan province using the standard microscopic agglutination test (MAT). For this purpose, 300 blood samples were collected from slaughtered cows in Guilan province during the spring and summer of 2023. After separating the serum and sending them to the leptospirosis research laboratory, all the samples were tested by MAT method using five serotypes of Leptospira Grippotyphosa, Pomona, Icterohaemorrhagiae, Canicola and Hardjo. The results of this study showed that 62 samples (20.66 percent) had anti-leptospiral antibodies against one of the above-mentioned 5 serotypes. The highest titer was related to Hardjo serotype with 37 samples (59.6 percent) and the lowest was related to Icterohaemorrhagiae serotype with 5 cases (8.06 percent). Titration of positive samples showed that 59 samples (86.8 percent) had a titer of 1:100 to 1:200 and 9 samples (13.2 percent) had a titer of 1:400. In conclusion, this study revealed that infection with L. interrogans is highly prevalent in slaughterhouse cows of Guilan province and the active presence of the cause of this disease in the province.            The aim of this study was to investigate changes of serum parameters such as adenosine deaminase (ADA), heat shock protein-27 (HSP-27), homocysteine (Hcy) and lipid profiles (LDL,HDL,VLDL and Triglyceride) in bovine leptospirosis. After diagnosis of acute leptospirosis in cattle, blood samples were collected from 12 cases of bovine leptospirosis and 12 healthy samples via jugular vein and all parameters along with zinc (Zn2+) were measured. The results denoted a significant increase in Hcy, HSP-27, TG, VLDL, along with a significant decrease in HDL-C, ADA, LDL-C and zinc (Zn2+) in patients compared to healthy ones (p≤0.01). Based on the results, the listed parameters may be used in the management of bovine leptospirosis. Manuscript profile

Leptospirosis  is  a  worldwide  bacterial  zoonoses  that  its  main  source  are  wild animals  and  rodents  That  leptospirals  excrete  in  their  urine. This  disease  causes  fever, hemoglobinuria, jaundice, abortion, mastitis, reduced  milk  production, reproductive  disorders  and  death  in  cattle, that  major  complications  in  sheep  are  abortion  and  pyelonephritis. The  purpose  of  this  study  is the  evaluation  of   risk  factors  for  leptospirosis  disease  and  its  association  with  high  prevalence  of  infection  in  the  sheep  flocks. Accordingly, 260 serum samples were collected from 20 flocks of  sheep. Then  with  microscopic  agglutination  test (MAT) using  6  live  antigen  Leptospira  interrogans  include  Pomona , canicola, icterohaemorrhagiae, gryppotyphoza, ballum  and  hardjo  was  studied. MAT  test  results  of  the  serum  samples  showed  the  contamination  was  28.46%. Most  contamination  of  the  samples  was  allocated  to  sheep  who  lived  in  swampy  areas, so  that  70.28%  of  positive   samples  were  included. 56.25%  of  positive  samples  among  the  samples  showed  positive  reaction  with  serotype canicola. On the other hand, 76.56%  of  the  positive  samples  belonged  to  animals  which  in  their  environment, rodents   in  very  large  numbers  were  observed. The  sheep  that  kept  in moist  and  wet  environment  to  be  had  60%  of  positive  samples , and  finally  the  highest frequency  of  positive  animals  were  ranged  among  ages  3  to  6  years  old, the  incidence  of  the  disease  increased  in  older  animals. By  attention  to  the  results, increasing  of  age, living  in  swampy  and  wet  bed, being  infectious  dogs  in  side  of  flocks and  also  very  large  number  of  rodents  in  environment  are  of  the  most  important  causes  of  seroprevalense  of  leptospirosis.   Manuscript profile

Leptospirosis is a zoonotic disease that can cause heavy economic losses through complications including abortion, stillbirth, infertility, reduced milk production, etc. In this study, 251 referred samples of aborted fetuses inindustrial dairy herds of Tehran province evaluated in MABNA laboratory during one year. Tested samples included a homogenous tissue mixture of fatal heart, kidney, liver and spleen. For DNA extraction, the ProteinaseK Method was used. Then polymerase chain reaction (PCR) was performed to recognize Leptospira Spp. by using specifc 16s rRNA gene primers. Samples with 331bp band in electrophoresis gel were considered positive.Prevalence of leptospiral abortion in average was 12.8 % in Tehran province during a year and based on season, respectively, the amount in spring was 15%, summer 9.08 %, autumn 10 % and winter 19.2 %. Based on theseresults, it was most prevalent in spring and winter that is probably due to higher environmental humidity, hydrophilic nature of leptospira and more bacterial activities. Generally according to the prevalence of 12.8 %, the majorcause of abortion in Tehran province in industrial dairy herds is leptospiral infection so that any activities in orderto eradicate and control the disease is essential.
