        Introduction:Nowadays infertility is recognized as one of the most common clinical problems around the globe, and one of the most worrying social issues in different cultures and societies. In the meantime, efforts have always been made to prevent the progression of infertility caused by this factor by carefully examining the most effective male factor - as one of the potential parties in infertility problems - that is, analyzing the quantity and quality of sperm. On one hand, traditional methods have lots of problems such as inadequate accuracy, clinicians' disagreements, and prolonged treatment. On the other hand, the successes of machine learning in many areas prompted researchers to move toward automating sperm morphology analysis by means of machine learning.Methods:The ladder network as a semi-supervised learning algorithm, by using a small number of labeled samples and a larger part of unlabeled data, shows suitability and compliance with the real-world requirements in this field of study. In this regard, in order to implement ladder networks, the structure of stack noise removal auto-encoders with the architecture of two parallel encoders has been used to represent the samples and a decoder to reconstruct the samples. The present study by applying changes and improving various factors, especially input noise, has obtained good results in the analysis of low-resolution images without coloring.Results:The proposed model succeeded by extracting positive and fruitful features from the images of the head, acrosome, and vacuole of human sperm, showing an acceptable accuracy for classifying them into two natural and abnormal classes, and finally selecting the appropriate sperm to participate in the artificial insemination process. The study of the proposed model for all three sperm sections (head, vacuole, and acrosome) succeeded, despite low-quality images, achieving impressive results of more than 70% for the head and acrosome and more than 80% for the vacuole.Conclusion: In the future, we intend to improve the proposed model by finding ways to increase the accuracy and reduce the error of the test results and show that the change in the type of noise or how it is applied to the network will have a significant impact on the network performance.         Inroduction and Objective: present study was aimed to evaluate the effects of addition of a synthetic antioxidant, MnTE, on motility and membrane integrity during the liquid storage (up to 72h) of goat semen.Materials and Methods:Semen samples were collected using artificial vagina from three mature goats, mixed and diluted (1:20 v/v) with a Bioxell extender containing 0 (control), 0.01, 0.1 and 1 µM of MnTE. All diluted sperm suspensions were cooled to 5°C and stored until 72hr and sperm motility and membrane integrity were assessed in 0, 12, 24, 36, 48 and 72hr post dilution.Results: With increasing storage time (0 to 72h), the percentage of motility and membrane integrity decreased in all treatments and control group. Addition of MnTE  at 0.01 and 0.1 µM concentration to the extender, improved goat semen quality, while addition of 1µM of MnTE had an adverse effect on motility and membrane integrity when compared to control.Conclusion: extender supplemented with MnTE could reduce the oxidative stress provoked by liquid storage processes. The optimum concentration of MnTE seems to be 0.01 and 0.1µM for goat semen in liquid storage state. However, further supplementation of extender with 1µM of MnTE not only did not improve the results but inversely affected motility and membrane integrity in all treatment group. Manuscript profile

Mobility percentage, pH, mobility duration, compaction and osmolarity of sperm which affect the quality of sperm were tested in this research. This study was carried out in April 2011 in Shahid Ansari Bony Fishes Propagation and Rearing Center. According to the results, mean and standard deviation of ions in 3 and 4 years old fish were 329.6±11.9 and 343.4±11.9 osmolarity, 47.2±7.4 and 44.5±2.99 spermatocrit, 60.5±12 and 66.3±6.1 mobility duration, 80±6.7 and 83.5±3.9 mobility percentage, 17828750±3199657.8 and 18000000±2090454.5 compaction of sperm and 7.79±0.51 and 7.61±0.26 pH of sperm.             The aim of this study was to investigate the effect of three concentration of citric acid on motility patterns of bovine epididymal sperms. For this purpose, 50 pairs of bovine testicles were collected immediately after slaughter form urmia abattoir and transferred to the laboratory alongside 5oc ice pack. Epididymal tail sperms were collected with a few incisions in vascular areas and transferred to hams f10 milieu with 10% fetal calf serum and counted after 15 minutes of incubation at 37oc in Co2 incubator. Concentrations of 50 million sperms per ml were proposal and in the normal sperm pH rang of 6.7-7.4, 0.1, 0.2 and 0.3 normal concentration of citric acid were added to sperm continuity micro tubes (normal concentration of acid equals 7 mg/ml of bovine serum) and at 15, 30, 45, 60, 90, 120, 180, 240 and 360 minutes the motility patterns of epididymal sperms were evaluated using the computer assisted sperms analyzing (CASA) method. Data were analyzed with one-way ANOVA using the SPSS 15 software. The results indicated significant differences in various indices of sperm motility patterns (Curvilinear   Velocity, Straight-line Velocity, Average Path Velocity, Mean Angel Degree, Amplitude of Lateral Head Displacement, Beat-Cross Frequency, Linearity, Wobble) particularly at 0.3 normal concentration of citric acid compared with the control.         در این مطالعه، تشکیل هیدروژن پراکسیداز (H2O2) توسط سرد کردن اسپرماتوزوآ بز اندازه‌ گیری شد. علاوه بر این، تأثیرات اسپرماتوزوآ مرده روی ویژگی‌های جنبایی مطالعه شدند. اجاکولیت‌های جمع‌آوری شده تازه از پنج بز نر شامی سانتریفیوژ شدند و تقریباً تمامی پلاسمای سمینال حذف شد. قسمتی از اسپرماتوزوآ جمع‌آوری شده توسط دو روش کشته شدند: ابتدا توسط فریز مکرر در نیتروژن مایع و یخ­ گشایی در 37 درجه سلسیوس در حمام آب و دوم توسط اضافه کردن اسپرماتوزوآ به آب دوبار تقطیر. در آزمایش اول، یک آزمون فلوسیتومتری با عامل 10-استیل3،7-دی‌هیدروکسیفنوکسازین به‌عنوان یک پروب برای تشخیص H2O2 برای اندازه‌ گیری تشکیل H2O2 استفاده شد. در آزمایش دوم، تأثیرات اضاقه کردن صفر (کنترل)، 25، 50 و 75 درصد (V/V) اسپرماتوزوآ مرده برای زندگی کردن جنبایی اسپرم با استفاده از آنالیز رایانه‌ای اسپرم (CASA) مورد ارزیابی قرار گرفتند. هیدروژن پراکسیداز از اسپرماتوزآ سرد شده زنده و مرده تولید شد و مقادیر این عامل با زمان افزایش داشت. علاوه بر این، تفاوت‌های معنی‌‌دار واضحی (0.05>P) بین سطوح تولید ­شده از اسپرماتوزوآ مرده در مقایسه با زنده مشاهده شد. اسپرماتوزوآ مرده توسط تیمار فریز و یخ‌گشایی مکرر مقدار بیشتری H2O2 تولید کرد (P>0.05). ارزش‌های درصد جنبایی اسپرماتوزوآ (MOT %)، درصد پیش‌رونده جنبایی اسپرماتوزوآ (PMOT %) و متوسط سرعت مسیر (VAP) به‌طور معنی‌داری کاهش داشتند (0.05>P) در مقایسه با کنترل‌ها زمانی‌که اسپرماتوزوا مرده اضافه شد. اثر منفی روی پارامترهای CASA قبلی افزایش داشت زمانی‌که درصدهای اسپرماتوزوآ مرده افزایش داشت هر آنچه که مسیر مرگ اسپرم بود. در نتیجه، تشکیل زیاد H2O2 از اسپرماتوزوآ سرد شده مرده بز ممکن است مسئول کاهش جنبایی باشد. حذف اسپرماتوزوآ مرده از محیط کشت انکوبه می‌تواند به بهبود ویژگی‌های جنبایی اسپرم سرد شده بز کمک نماید.         اثرات پنج سطح اسمولاریته و pH همراه با سه دمای مختلف روی پارامترهای تحرک اسپرم قوچ­های آواسی با استفاده از تحلیل­گر اسپرم مبتنی بر کامپیوتر (CASA) در محیط کشت­های تیرود آلبومین لاکتات پیرووات (TALP) و تریس زرده تخم مرغ (YET) مطالعه گردید. اسپرم حاصل از 5 قوچ آواسی بالغ با هم مخلوط گردیده و روی دو محیط با میلی­اسمولاریته 100، 200، 300، 400 و 500 به ازای هر کیلوگرم در دمای 4، 20 و 37 درجه سلسیوس و نیز در سطح pH 5، 6، 7، 8 و 9 در دماهای یکسان رقیق گردیدند. اسمولاریته، pH و دما اثر معنی­داری (05/0>P) روی درصد تحرک (MOT %) و درصد اسپرم با حرکت پیش­رونده (PMOT %) داشتند. غیر از ناهنجاری سر جانبی، همه پارامترهای دیگر تحرک CASA نیز به طور معنی­داری تحت تأثیر قرار گرفتند (05/0>P). اسپرم انکوباسیون شده در YET قادر بود در اسمولاریته­های بین 200 تا 400 میلی­اسمول به ازای هر کیلوگرم دامنه برای MOT % مقاومت کند. شرایط بازی در 9 =pH اثر منفی بزرگتری در مقایسه با شرایط اسیدی در 5 =pH داشت. در مقایسه با 20 و 37 درجه سلسیوس، دمای 4 درجه سلسیوس تأثیر منفی بر MOT % و PMOT % داشته و این امر در محیط TALP با وضوح بیشتری دیده می­شد. به عنوان نتیجه­گیری، نتایج ما نشان داد که: 1) اثرات اسمولاریته، pH و دما بر اسپرم آواسی انکوباسیون شده در هر دو محیط TALP و YET معنی‌دار بود و 2) لازم است که دما برای اسپرمی که در تلقیح مصنوعی و آزمایشات باروری خارج رحمی در این دو محیط مورد استفاده قرار می­گیرند، به دقت انتخاب شوند.         گیاه مرزنجوش دارای سطوح بالایی از ترکیبات فنولی نظیر اسید گالیک و فنل­هایی نظیر اسید رزمارینیک و کوئرسیتین است. هدف از مطالعه حاضر بررسی اثرات عصاره مرزنجوش به عنوان آنتی­اکسیدان طبیعی، بر روی کیفیت منی منجمد-یخ­گشایی شده گاو نر هلشتاین بود. اسپرم­گیری دو بار در هفته، به مدت دو ماه و از سه گاو نر هلشتاین (6-5 ساله با میانگین وزنی 800 کیلوگرم) انجام گرفت. عصاره اتانولی مرزنجوش در مقادیر 20، 16، 12، 8، 4، 2 میلی­لیتر در دسی­لیتر رقیق­کننده به­رقیق کننده بر پایه سیترات-زرده تخم مرغ افزوده شد. بعد از یخ­گشایی نمونه­های منی، پارامترهای تحرکی اسپرم، زنده­مانی و یکپارچگی غشاء به ترتیب توسط سیستم کاسا، رنگ آمیزی ائوزین-نیگروزین و تست التهاب هیپواسمتیک مورد ارزیابی قرار گرفتند. همچنین مقادیر مالون دی آلدهاید و فعالیت سوپراکسید دیسموتاز و کاتالاز اندازه­گیری شدند. نتایج بیانگر بالا بودن درصد تحرک (98/7±34/72) در نمونه دریافت کننده 4 میلی­لیتر در دسی­لیتر عصاره مرزنجوش بود (05/0>P). افزودن 2 و 4 میلی­لیتر در دسی­لیتر عصاره مرزنجوش باعث بهبود معنی­دار میزان تحرک، یکپارچگی غشاء پلاسمایی اسپرم­ها بعد از فرآیند انجماد-یخ­گشایی نسبت به گروه شاهد شد. افزودن 4 میلی­لیتر در دسی­لیتر عصاره مرزنجوش باعث کاهش غلظت مالون دی آلدهاید نسبت به گروه شاهد شد (05/0>P). میزان فعالیت سوپراکسید دیسموتاز و کاتالاز در نمونه­های دریافت کننده 8 و 4 میلی­لیتر در دسی­لیتر عصاره مرزنجوش به طور معنی­داری افزایش یافت (به ترتیب 18/0±86/1 و 28/0±92/1 واحد بر میلی­گرم پروتئین؛ 13/0±54/4 و 28/0±28/4 واحد بر میلی­گرم پروتئین) (05/0>P). میزان فعالیت سوپراکسید دیسموتاز و کاتالاز به طور معنی­داری در نمونه­های دریافت کننده 4 و 8 میلی­لیتر در دسی­لیتر عصاره مرزنجوش افزایش یافت (05/0>P). به طور کلی، افزودن 2 و 4 میلی­لیتر در دسی­لیتر عصاره مرزنجوش به رقیق­کننده منی باعث بهبود کیفیت منی بعد از انجماد-یخ­گشایی ­شد که ممکن است به دلیل افزایش فعالیت آنزیم­های آنتی­اکسیدانی و کاهش پراکسیداسیون لیپیدی باشد.         Semen analysis is one of the most important methods that reflects the fertility potential of men. Many factors lead to infertility in men, among which urogenital bacterial infections seem to play an effective role. Therefore, this study was planned to evaluate the frequency of bacterial infection in men with different infertility factor. Then, the effect of bacteriospermia was studied on the basic semen parameters and assisted reproductive outcomes.In this study, 98 semen samples from infertile men with male-factor infertility were collected. The semen samples were analyzed based on World Health Organization (WHO) criteria and categorized to four groups with male factor infertility. About 0.5-1 mL samples were prepared and used to inject into oocytes. To evaluate bacterial infection, the remaining samples were transported to microbiological laboratory during 1 hour to culture using standard bacteriological techniques. The bacterial infections such as E. coli (12.24%) and Staphylococcus saprophyticus (28.57%) were detected in 40.81% of the cultured samples. The basic semen parameters such as concentration, progressive motility, viability (p<0.05), and normal morphology (p<0.01) of sperms were decreased in the samples with bacterial infections. The clinical pregnancy was also decreased in the bacteriospermia groups (p<0.05).         AbstractIntroduction : Pre-sex selection is the process by which Y or X-bearing spermatozoa are used to produce the desired sex in the offspring. X-bearing spermatozoa are believed to have a longer lifespan, and are more resistant to acidic conditions, temperature changes, and even oxidative stress compared to Y-bearing. Methods : The present study was aimed to investigate the effect of heat stress on sperm parameters and sex ratio of sperm containing X or Y chromosome after freezing-thawing of goat semen. The diluted semen of the heat stress group was first placed at 43 ° C for 30 minutes and then frozen-thawed as in the control group. Results : The results showed that heat stress led to a significant reduction in the overall sperm motility, viability and sperm cell membrane integrity compared to the control group. Also, in the heat stress group, the mean Y to X ratio was 94.4%, which showed a significant difference (p <001) compared to the expected Y to X ratio of 1.Conclusion : Applying heat stress to sperm in vitro can lead to a significant reduction in sperm parameters.         The maltose disaccharide is easily converted into two glucose molecules during cellular glycolysis and is an energy source for sperm motility. To evaluate the effect of maltose on the kinetics of water buffalo epididymal sperm, 10 pairs of buffalo bull testicles, after the usual industrial slaughter, were transported to the laboratory of the Faculty of Veterinary Medicine in a polystyrene box at a temperature of 5o to 8o Celsius. In the laboratory, after fixing the tail of the epididymis with two fingers, several incisions were made in noncapillary zones. After the milky liquid containing concentrated spermatozoa was extracted, this liquid was transferred to an Eppendorf cell culture medium containing 10% bovine serum albumin (BSA). Five levels of maltose sugar (1, 3, 5, 10, 15 mM) were added to the Eppendorf containing 1 ml culture medium and 30-40 million sperm with 10% BSA and incubated at 37o C for 24 hours. Sperm kinetics were assessed at 1, 6, 12 and 24 hours by computer-assisted sperm analysis (CASA). Statistical analysis showed that CASA data such as rapid progressive motility (class A, %), progressive motility (class B, %), viability (class A+B+C, %), straight-line velocity (VSL, µm/s), curvilinear velocity (VCL, µm/s), average path velocity (VAP, µm/s) and lateral head amplitude (ALH, µm) at 24 hrs were higher in the maltose group than in the control group (p<0.05). In conclusion, this study suggests that maltose has a desirable effect on buffalo epididymal spermatozoa and maltose may act as an energy substrate for sperm motility. Manuscript profile

        Polystyrene microplastics (PS-MPs) pose a significant scientific concern due to their chemical makeup. Luteolin (LUT), a phytochemical-biological substance with anti-oxidative and anti-inflammatory properties, was tested against polystyrene microplastics (PS-MPs)-induced reproductive toxicity in male Wistar rats. The rats were randomly assigned to 4 groups and treated as follows: Group I received 0.01 mL kg-1 of DMSO alone, Group II received 100 mg kg-1 of LUT dissolved in DMSO, Group III received 0.01 mg kg-1 of PS-MPs, and Group IV received 0.01 mg kg-1 of PS-MPs and 100 mg kg-1 of LUT. PS-MPs alone dramatically increased sperm morphological defects, xanthine oxidase, and lipid peroxidation while decreasing testicular enzymes, luteinizing hormone (LH), follicle-stimulating hormone (FSH), testosterone, antioxidants, and interleukin-10 (IL-10). The study demonstrates that PS-MPs can alter male rat testicular function and androgenicity. However, as a result of LUT co-treatment, sperm count, motility, and viability were increased whereas MDA and XO, IL-1β and TNF-α, and caspase-3 activities in the testis were reduced. Supplementing with LUT might act as a cytoprotective agent to lessen the spermatotoxicity and dysgonadogenesis brought on by PS-MPs. Manuscript profile

        Aloe with the scientific name Aloe vera is a perennial plant that has useful effects in herbal therapy and herbal medicine. In this research the effects of hydroalcoholic extract of Aloe vera gel on the testis and spermatogenesis in mice Balb/C mice were examined. Samples randomly divided into five groups (three treatment groups, placebo and control) and all samples were kept in the same conditions. Aloe vera gel extract in different doses (50, 100 and 200 mg/kg/2day) produced by intraperitoneal (IP) for 20 days was administered to the treatment group. Placebo group was used as the injection of saline. The main parameters that were examined changes in levels of testosterone, LH and FSH levels and changes in stem cell number, number primary spermatocytes, and testis weight compared with the control group. All of hormonal experiments using Radioimmunoassay (RIA) were performed.The results showed that Aloe vera extract caused a significant increase in the number of stem cells and primary spermatocytes in the experimental group compared with the control group. Mean of testicular weights and testosterone levels in group 3 (200 mg/kg) increased significantly, while LH and FSH levels did not show significant changes. The findings indicate Aloe vera extract in a dose-dependent effect on testosterone levels and histological characteristics of male mice. Manuscript profile

The aim of this study was to investigate the effect of various concentrations of ginger extract Specific parameters of sperm in roster. In this study, hydroalcoholic extracts of knotgrass was prepared in Golpaygani roosters. In this study, hydro-alcoholic extract of ginger plant was prepared at concentrations of 0, 500, 1000 and 2000 mg/L and was added to drinking water of 36 adult male (32 weeks old) Golpaygani cock. After two week, the effect of different concentrations of the extract on the specific indices of sperm fertility (SMI, FSC, PMSCa, PMSCb and MSC) were evaluated and compared. The papenicular staining was also used to specific evaluation of sperm abnormalities. The use of ginger extract significantly improved the sperm fertility indices. The most number of sperm with fast progressive movements (PMSCa), the highest levels of SMI, FSC and MSC indices, the least structural and motility abnormalities was demonstrated in 1000 and 2000 mg extract (P≤0.05). Significant differences were not observed between the two above-mentioned intermediate and high concentrations in some parameters (P> 0.05). There was no significant difference in above indices between medium and high concentrations of extract . In conclusion, ginger , due to the high amounts of antioxidant compounds, significantly increases the sperm indices and also improves the specific parameters of sperm fertility in rooster.         Good semen quality plays a very important role in promoting poultry breeding programs; due to its antioxi-dant properties and role in fat metabolism, L-carnitine seems to be effective in improving semen quality. Therefore, this study was performed to evaluate the effect of dietary L-carnitine supplementation on the motility parameters and qualitative traits in semen Cobb breed roosters. A total of twenty roosters, 24 weeks-old Cobb roosters randomly assigned to four experimental groups. The experimental design was conducted in a completely randomized design with 4 experimental treatments (T1: 0, T2: 125, T3: 250, and T4: 500 mg L-carnitine per kg diet) with 5 replications and sampling twice (semen collection) with an in-terval of 14 days. The collected semen was immediately evaluated for motility parameters and sperm qual-ity traits. The results showed that there is no significant difference between the levels of 250 and 500 mg of L-carnitine in semen parameters, but there are significant differences with other groups (P≤0.05). Semen collection time did not affect sperm parameters (P≤0.05). In conclusion, the addition of L-carnitine at a rate of 250 mg/kg diet can improve motility parameters and sperm quality traits in the Cobb rooster. Manuscript profile

        Carob belongs to the Fabaceae family, which has been studied for its medicinal properties, showcasing an-tioxidative properties and the ability to enhance sexual and immune system functions. This study aimed at examining the effect of adding hydro-alcoholic extracts of carob seeds to the diet on steroid hormones, sperm parameters and blood parameters of Lori-Bakhtiari rams. In this study, carob seeds were extracted using the percolation method. Thirty Lori-Bakhtiari rams were selected with a mean weight of 50 ± 5 kg and mean age of 2.5 years old. They were equally divided into three groups: (1) control (without carob ex-tract), treatment (1) 150 mg/kg body weight (BW), and treatment (2) 300 mg/kg of carob seed extract in the diet for 4 weeks (2021-2022) for adaption process. Then semen and blood were analyzed for four months. Sperm motility, viability and the hypo-osmotic swelling (HOS) tests were evaluated at pre-freezing and post-thawing by the Computer Assisted Semen Analysis (CASA) software, Eosin-Nigrosin staining and a phase-contrast microscope, respectively. Furthermore, blood samples were collected for evaluation of fast-ing blood sugar (FBS), cholesterol (CH) and triglyceride (TG), serum creatinine (sCr), high-density lipopro-tein (HDL), low-density lipoprotein (LDL), and testosterone hormone (T). The results of this study showed that the sperm parameters like motility and viability in the treatment groups increased significantly (P<0.05) in pre-freezing and post-thawing evaluations. Additionally, the results of blood parameters showed that FBS, CH, TG, LDL and sCr were significantly decreased by using the carob extract in a diet (P<0.05), while the testosterone level significantly increased during the experiment (P<0.05). In conclusion, the use of 300 mg/kg carob extract in the diet showed a more positive effect on sperm and blood parameters at Lori-Bakhtiari rams. Manuscript profile
