Approaches Pertaining to Cryopreservation in Stem Cells and Cancer Stem Cells; Impact of Cryobanking on Surface Characteristics of Mammospheres Utilizing Scanning Electron Microscopy

Abstract :
The importance of cryopreservation and biobanking of stem cells has significantly increased owing to their applications across diverse domains. Stem cells are currently employed in the treatment of some diseases and are anticipated to play a crucial role in the future of medicine. On the other hand, cancer stem cells (CSCs) have emerged as potential therapeutic targets, thereby profoundly transforming paradigms within cancer research. Consequently, the long-term cryopreservation of CSCs is of paramount importance. Nonetheless, existing evidence suggests inconsistent results concerning the effects of cryopreservation and thawing on stem cells, which include various forms of cellular stress and damage. Moreover, there is documentation of changes in biological characteristics and integrity in stem cells following thawing. Mammospheres derived from 4T1 cell lines were frozen in standard freezing medium (90% FBS+10% DMSO) and stored in liquid nitrogen for 9 months. Then, cryopreservation effect on preservation of CSCs maintaining the integrity of cancer stem cells was evaluated using electron microscopy and compared with their fresh counterparts. The data obtained from scanning electron microscopy micrographs unequivocally demonstrated that cancer stem cell-enriched mammospheres preserved their structural integrity, morphological characteristics, and cell surface properties following the thawing process. Consequently, this cryopreservation technique offers researchers a robust and dependable approach for the conservation of cancer stem cells for prospective research endeavors.
