In this study the effect of DNA concentration and incubationtime of sheep spermatozoa with foreign DNA on DNA uptake, its dynamic and spermfunctionality and their difference between ejaculated and epididimal sperm was investigated.For this the ejaculated(N=4) and epididimal(N=6)spermatozoa was incubated with differentconcentration (5-500ng/106 sperm/25μl) of rhodamine labaled pEGFP-IRES-hLys in varioustime (15-90min) and DNA uptake, dynamic of uptake and sperm functionality was assayed.Results showed to an average of 65% of incubated spermatozoa uptake the foreign DNA,depend on DNA concentration and incubation time. Although the amount of transfectedspermatozoa could increase significantly with prolonged incubation time up to 60 min but theamount of DNA uptake was higher in the first 15min of incubation. Increase of foreign DNAconcentration and incubation time increased the transfected spermatozoa and uptake intensity. Itseems higher DNA concentration, more rapid DNA uptake and more rapid gain of transfectionpeak. Slight decrease of sperm total and progressive motility was seen in treatment group incomparison with Control groups (except in 5min incubation). No spermatozoa with postacrosomal absorption (true uptake) were motile. So incubation for more than 30 and 60 min inmoderate (20-200ng) and low (5ng) plasmid concentration respectively is not useful. The purpose of this research was to glycosylate lysozyme with dextran through Maillard-based reaction and to study the antimicrobial characteristics of lysozyme-dextran conjugate.Materials and Methods: Glycosylation of lysozyme with dextran was performed using a 1:5 weight ratio of protein to dextran, incubated at 60 MC for one week under the relative humidity of 79%. For evaluating conjugation of dextran to lysozyme, SDS-PAGE electrophoresis was conducted. Gel filtration chromatography with sephadox G-100 was used for separation of conjugated enzyme. Lytic activity, free amino group and antimicrobial activity of the modified enzyme were evaluated.Results: SDS-PAGE electrophoresis was used to follow the glycosylation process. Results indicated that 3.7 moles of dextran were coupled to one mole of lysozyme. The lytic activity of the conjugate was about 62% of that of the native lysozyme. Evaluation of antimicrobial activity of the lysozymedextran conjugate, indicated the effectiveness of modified enzyme against E. coli and a progressive increase in antimicrobial activity with an increase in enzyme-conjugate concentration. The antimicrobial action of lysozyme on S. aureus was not improved by conjugation with dextran as compared with that of E. coli.Conclusion: These results might increase the application of lysozyme as a natural antimicrobial ingredient in different food systems. The aim of this study was to investigate the effect of high-speed sports activity on the anti-inflammatory factors lysozyme, LL-37 and HDB-2 in the saliva of obese adolescent boys. Materials &Methods: 32 male students voluntarily participated in the present study and were randomly assigned to four groups: 1) obese aerobic exercise, 2) normal weight aerobic exercise, 3) obese control without exercise, and 4) normal weight control without exercise. Anthropometric indices of height, weight and body mass index were measured. Before and after eight weeks of practice, Shatell-Run standard test was performed. Salivary samples of lysozyme, lactoferrin, lactate and C-reactive protein concentrations were taken after eight weeks of training with a frequency of three sessions per week with an intensity of 30 to 90% of maximum aerobic power. Using analysis of covariance, variables with a significance level of less than (p≥0.05) were included in the analysis. Results: The results showed that the salivary levels of lysozyme (p=0.001), LL-37 (p=0.002), and HDB2 (p=0.001) increased significantly, and the amount of increase in obese students was higher than in people with normal weight. Conclusion: The increased response of some salivary anti-inflammatory proteins following eight weeks of high-speed exercise training after increased activity may be due to the short-term responses of the immune system against the pressures caused by intense activity. Chicken fillets were treated in whey protein isolate and whey protein isolate containing 0.5, 1 % Lysozyme and compared with chicken fillets without any coating (control) with 3 replicates were prepared. The microbial parameters (Total bacterial Mesophilic, Enterobacteriaceae, Psychrotroph and Pseudomonas spp count) were evaluated for 12 days. Results showed that during the storage time, in the samples coated with whey protein isolate containing different concentration of Lysozyme, a significant reduction (p< 0.05) were observed in the entire evaluated microorganism groups compared to the control samples. Also a dose related trend was observed due to addition of Lysozyme. Overall the findings of present study suggest that whey protein isolate contain with Lysozyme, may use as a natural coating and preservative to extend the chicken meat shelf life. This study amid to verify the effect of lysozyme concentration on Streptococcus iniae and Lactococcus bacteria .It also determined minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of these compounds on the growth of mentioned bacteria. Present study investigated antimicrobial effect of different concentrations of Lysozyme (5000, 2500, 1250, 625, 312.5, 156.25, 78.13, 89.06 and 19.53 µg/mL) against Streptoccus iniae and Lactococcus garviea isolated from rainbow trout in Chaharmahal-o-Bakhtiary province. The study used the methods of light (optical) absorption and microdilution in three pH including 5.5, 6, 7, as well. The result of microdilution showed the effect of lysozym in the inhibition of the growth of both bacteria at pH 5.5 and 6 on concentration of 5000 µg/ml, whereas, at pH7 was observed any inhibition. The results showed declining light absorption values resulting from the growth of both bacteria with increasing concentration of lysozyme, however the inhibition effect of 5000 µg/ml of lysosyme at pH6 and 5.5 was significantly (p<0.05) less, in comparison with the smaller concentrations. On the other hand, any inhibitory effect as the optical absorption was observed in the growth of bacteria for the highest concentration of lysozyme in the media of pH 7. Manuscript profile -

8 - تغذیه خانوادههای زنبور عسل با یک افزودنی سولفات کبالت
ر. بالکانسکا د. سالکووازنبورهای عسل (Apis melifera) برای زندگی معمول خود نیازمند پروتئین­ها، کربوهیدرات­ها، لیپیدها، ویتامین­ها و مواد معدنی هستند. به خوبی مشخص شده است که کبالت و ویتامین B12 دو ماده مغذی هستند که در گرده و گیاهانی که تأثیر مثبتی در تغذیه زنبورهای عسل داشتند، گزار Moreزنبورهای عسل (Apis melifera) برای زندگی معمول خود نیازمند پروتئین­ها، کربوهیدرات­ها، لیپیدها، ویتامین­ها و مواد معدنی هستند. به خوبی مشخص شده است که کبالت و ویتامین B12 دو ماده مغذی هستند که در گرده و گیاهانی که تأثیر مثبتی در تغذیه زنبورهای عسل داشتند، گزارش گردیده­اند. بدین منظور، هدف از این مطالعه تعیین تأثیر تغذیه خانواده­های زنبور عسل با سولفات کبالت به عنوان یک مکمل، بر ترکیب شیمیایی بدن زنبورهای کارگر و برخی از تولیدات زنبور عسل بوده است. این تحقیق در طی ماه­های می تا آگوست 2015 در بخش تحقیقات زنبور عسل موسسه علوم دامی کاستینبراد انجام دشه است. 6 خانواده زنبور عسل در قالب 3 خانواده آزمایشی (تغذیه شده با شربت شکر، شکر به آب با نسبت 1 به 1 و 4 میلی­گرم در هر لیتر سولفات کبالت به عنوان یک مکمل) و 3 خانواده شاهد (تغذیه شده فقط با شربت شکر) مورد مطالعه قرار گرفتند. نتایج نشان داد که افزودن 4 میلی­گرم در هر لیتر سولفات کبالت در شربت شکر خانواده­های زنبور عسل تأثیری بر میانگین وزن و ترکیب شیمیایی زنبورهای کارگر ندارد. تفاوت­های معنی­دار آماری (05/0>P) در فعالیت دیاستاز عسل در گروه خانواده­های آزمایشی دیده شد. 9 - احتمالاتی برای بهبود ایمنی ذاتی هومورال در بوقلمونها و مرغها در شرایط استرس دمایی توسط تعدیلکننده سیستم ایمنی ®Immunobeta
ن. بوزاکووا ل. سوتیرو س. دنو ت. کوینارسکیآزمایشهایی برای ارزیابی اثر Immunobeta با بوقلمونها و مرغهای تخمگذار پرورش­یافته ارگانیک انجام شد. بوقلمونها Immunobeta را در یک دز چهار گرم/کیلوگرم خوراک در طول شروع تخمگذاری (دوره سرد)، دوره حرارتی خنثی و دوره گرم تابستان دریافت کردند. در مرغها، دو سطح جیرها Moreآزمایشهایی برای ارزیابی اثر Immunobeta با بوقلمونها و مرغهای تخمگذار پرورش­یافته ارگانیک انجام شد. بوقلمونها Immunobeta را در یک دز چهار گرم/کیلوگرم خوراک در طول شروع تخمگذاری (دوره سرد)، دوره حرارتی خنثی و دوره گرم تابستان دریافت کردند. در مرغها، دو سطح جیرهای Immunobeta آزمون شدند: دو گرم/کیلوگرم و چهار گرم/کیلوگرم. سرم خون و سطوح لیزوزیم سفیده تخم مرغ، مسیر جایگزین فعال­سازی مکمل (APCA) و غلظتهای بتا-لایزین تعیین شدند. مشخص شد که Immunobeta تأثیر سودمندی بر لیزوزیم سرم، فعالیت APCA و بتا-لایزین در بوقلمونها و مرغها داشت. غلظت لیزوزیم در سفیده تخم مرغ به طور معنیداری در تخمهای تولید شده توسط پرندگان تیمار شده با هر دو دز تعدیل­کننده سیستم ایمنی بالاتر بود. Immunobeta، پیشنهاد شده با یک دز چهار گرم/کیلوگرم خوراک تأثیر سودمندی روی فاکتورهای ایمنی طبیعی هومورال داشت. با اعمال یک دز چهار گرم/کیلوگرم، تعدیل­کننده سیستم ایمنی تست شده به طور معنیداری غلظتهای لیزوزیم سفیده تخم مرغ را در مرغها افزایش داد، با فرض ماندگاری بالاتر تخمها و احتمالاً، محافظت بهتر از جنین جوجهها. س. لازاروو پ. ولوا آی. ژلیازکووارفتار بهداشتی 20 کلنی زنبور عسل در مناطق مختلف بلغارستان آزمون شد. درجه بیان رفتار بهداشتی توسط روش pin-killing ارزیابی شد. یک آزمون مزرعهای مربع 5 × 5 سانتی­متری نصب­شده روی بخشی از شان عسل با یک بچه کارگر چسبیده انجام شد. کلنیهای زنبور به دو گروه تقسیم Moreرفتار بهداشتی 20 کلنی زنبور عسل در مناطق مختلف بلغارستان آزمون شد. درجه بیان رفتار بهداشتی توسط روش pin-killing ارزیابی شد. یک آزمون مزرعهای مربع 5 × 5 سانتی­متری نصب­شده روی بخشی از شان عسل با یک بچه کارگر چسبیده انجام شد. کلنیهای زنبور به دو گروه تقسیم شدند: بهداشتی (پاک کردن بیش از 95 درصد سلولها در منطقه مورد آزمون در طی 48 ساعت) و غیربهداشتی (تمیز کردن کمتر از 95 درصد سلول­ها در منطقه مورد آزمون در 48 ساعت). همولنف از زنبورها از هر کلنی زنبور در 48 ساعت گرفته شد و سطوح لیزوزیم و محتوای پروتئین تام تعریف شد. آنالیز چندمتغییره (ANOVA) برای تعیین تفاوت معنیدار بین کلنیهای بهداشتی و غیربهداشتی استفاده شد. نتایج به­دست آمده تفاوتهای معنیداری بین دو گروه (P<0.05) براساس درصد سلولهای تمیز شده بعد از کشتن بچه در ساعتهای 3، 24 و 48 و تفاوتهای معنیدار بین کلنیهای بهداشتی و غیربهداشتی (P<0.05) برای هر دو پارامتر لیزوزیم و پروتئین تام نشان داد. همبستگی دو ­متغییره برای بررسی تأثیر رفتار بهداشتی روی سطوح لیزوزیم و محتوای پروتئین تام بکار گرفته شد. همبستگی ژنتیکی منفی (r=-0.33) بین پروتئین تام و رفتار بهداشتی بدست آمد که به معنی آن است که افزایش فعالیتهای مربوط به پاکسازی سلولهای شانه زنبوری منجر به کاهش محتوی پروتئین تام میشود. همبستگی پایین (r=0.35) بین رفتار بهداشتی و سطوح لیزوزیم یافت شد بدین معنی که سطوح بالاتر لیزوزیم با رفتار بهداشتی بارزتری گزارش شده است. مدل رگرسیونی محاسبه شده برای لیزوزیم (P=0.24826) به لحاظ آمتری غیرمعنیدار است اما مدل رگرسیونی برای پروتئین تام به طور آماری معنیدار است (P=0.003153) و میتواند برای پیشبینی ارتباط بین محتوی پروتئین تام در همولنف و رفتار بهداشتی کلنیهای زنبور استفاده شود. Totally 360 Rainbow trout specimens (75±10g )were divided in four groups and were fed with diets containing 0, 5, 7.5 and 10 g of pure dried Sargassum in each kilogram of food respectively, for a period of two months. At the end of the second month of culture, blood samples were taken from 12 fish (3 fish from each group), and sent to the laboratory for analysis. Results indicated that levels of IgM and lysozyme activity in the blood plasma of fish fed with Sargassum illicifolium showed significant statistical differences (P<0.05) with control group. The obtained results, indicate that Sargassum, containing beta carotene, has favorable effects on immunology, resistance and growth of rainbow trout. Manuscript profile -
