Organ Specific Expression of Human Calcitonin Gene in Transgenic Potato plant

Abstract :
Molecular farming as a new and promising industry involvs plant biotechnology for the production of valuable pharmaceutical proteins in plants. Transgenic plants offer many advantages as low-cost system for large-scale production. The ability to carry out post translational modification similar to animal cells, the absence of human pathogens and low production costs are some of the advantages of plants. Among bioactive peptide, human calcitonin (hCT) hormone plays an important role for the treatment of disorders of bone and mineral metabolism, so its production in different hosts is attractive. The aim of this project was to investigate the organ specific expression of hCT gene in transgenic potato tuber. In this way, the expression cassette containing the synthetic hCT gene with optimized codon for plant expression under the control of class I Patatin promoter and Nos terminator was cloned in plant binary vector. The chimerical construct was integrated into potato plant chromosome by Agrobacterium mediated transformation. After regeneration, transgenic potato plants analyzed for the sense of hCT gene with PCR and the organ specific expression of human calcitonin were determind by ELISA . The highest level of expression was see with 1.5 ng protein per total soluble protein in tubers.
