        Insulin like growth factor-1 (IGF-I) has an antioxidant effect to maintain sperm motility. The present study designed to evaluate the effect of different levels IGF-1 on semen characteristic after cryopreservation of buck semen. Fifty ejaculates were collected and were extended with tris- citric acid- fructose diluent with 10% egg yolk and 6% glycerol with sperm concentrations of 4 × 108 mL−1. The effect of IGF-1 concentration showed significant differences (P<0.05) on most of studied traits (sperm motility, live sperm count, acrosomes integrity, hypo osmotic swelling positive spermatozoa, malondialdehyde (MDA)) and were significantly (P<0.05) highest in 250 ng/mL of IGF-1in post thaw semen. The post thaw progressive motility, live, hypo osmotic swelled and acrosome intact sperm increased by 25.26%, 26.77%, 30.35% and 25.52%, respectively in 250 ng/mL IGF1supplemented group in compare with control group. The 250 ng/mL IGF-1 supplemented group showed the lowest MDA content (P<0.05) in compare with other studied groups. The fortification 250 ng/mL IGF-1 reduced the MDA content by 31.16% more than the control group. The results suggested that IGF-1 protects sperm membrane lipid peroxidation by lowering MDA production by reducing the harmful effect of reactive oxygen species. Eight out of ten goats were conceived and kidded by using supplemented of 250 ng/mL IGF-1 and three goats were kidded by non-supplemented (control) frozen semen straw through artificial insemination (AI). The success of kidding was 40.00% in supplemented group (250 ng/mL IGF-1) than 30.00% in non-supplemented group (control) on actual kidding basis. پرونده مقاله

        This experiment was designed to study the effect of insulin like growth factor-I (IGF-I) on cryopreservation of Hariana bull semen. For this purpose, semen ejaculates were collected from four Hariana bulls using artificial vagina at biweekly interval. The semen samples which possess more than 70% initial progressive motility and above 500 million/mL spermatozoa concentration was subsequently subjected to processing for experiment. Semen samples were extended in egg yolk tris glycerol (EYTG) extender and split into four groups. IGF-I was added at 50 ng/80 × 106 spermatozoa, 100 ng/80 × 106 spermatozoa and 150 ng/80 × 106 spermatozoa in the group II, III, IV respectively and group I as control (without IGF-I). Semen samples were evaluated at pre-freeze and post-thaw stage for percent individual progressive motility, percent live spermatozoa, percent hypo-osmotic swelling (HOS) positive spermatozoa, percent spermatozoa with intact acrosome. A significant (p < 0.05) higher individual progressive motility, viability, HOS response and acrosomal integrity was observed at pre-freeze and post-thaw stage by using 150 ng/ml IGF-I. Concentration of 150 ng/mL IGF-I was found to be more beneficial in cryopreservation of Hariana bull spermatozoa as evidenced by post-thaw seminal parameter.         3 - Effect of Single Nucleotide Polymorphisms in <i>IGF-1R</i> Gene on Growth Rate Traits in Makooei Sheep

        Insulin-like growth factor 1 receptor (IGF-1R) is a main receptor of IGFs family which plays a critical role in the postnatal growth and skeletal growth in many species. However, there are few reports of IGF-1R gene structure and its effects on growth traits in sheep. The objectives of this study were detection of IGF-1R polymorphisms and assessment of their associations with growth traits in Iranian Makooei Sheep. Hence, 200 Makooei lambs were genotyped through polymerase chain reaction-single-strand conformation polymorphism (PCR-SSCP). The studied traits were birth weight (BW), weaning weight (WW), 6 months weight (6MW), average daily gains from birth to 3 months (ADG0-3), from 3 months to 6 months (ADG3-6), from birth to 6 months (ADG0-6) and corresponding Kleiber ratios (KR0-3, KR3-6, KR0-6).For this genetic position, three types of banding patterns (AA, AB and BB) were identified with the frequencies of 0.69, 0.16 and 0.15, respectively. In this study, IGF-1R genotypes indicated the significant associations with 6MW, ADG0-6, KR0-3 (P&lt;0.05) and ADG0-3 (P&lt;0.01). In all of the significant traits, The AAgenotype was linked to the highest values, while the BBgenotype was linked to the lowest values. The results of this study indicated that single nucleotide polymorphism (SNP) variation in IGF-1R gene can be used as a molecular marker for improving of growth traits in marker assisted selection programs in sheep. پرونده مقاله

        This experiment was conducted to investigate the effect of pre-pubertal plane of nutrition on the skeletal growth, lamb mortality, insulin-like growth factor1 (IGF-1) concentrations, quantity and quality of colostrum produced in ewe lambs. A total of 40 clinically health Kurdish female lambs (30&plusmn;8.6 d and weighing 10.2&plusmn;3.4 kg) were randomly allocated to one of two experimental diets in pre-weaning period: high quality diet (HQD, 2.50 Mcal ME/kg dry matter (DM) and 148 g CP/kg DM) or low quality diet (LQD, 2.02 Mcal ME/kg DM and 87 g CP/kg DM). At weaning, one half of lambs from each group was randomly separated and assigned to HQD or LQD. So there were four treatment groups in post-weaning period: H-H (HQD pre- and post-weaning); H-L (HQD pre-weaning and LQD post-weaning); L-H (LQD pre-weaning and HQD post-weaning) and L-L (LQD pre and post-weaning, control group). Weekly DM intake was determined. Serum IGF-1 concentrations was determined by ELISA method. The HQD treatment increased DM intake and BW (body Weight) compared with the LQD treatment during pre-weaning period(P&lt;0.01). At 210 d of age, animals fed LQD during the pre-weaning period and HQD during the post-weaning period (L-H sequence) had greater body length (BL), wither height (WH), hip height (HH) and hip width (HW) than animals on the H-L sequence. No interaction of the two periods was detected for heart girth, HW, WH, BL at 210 d of age (P&gt;0.05). HW at weaning time was not affected by quality of diet. However, lambs of H-H and L-H sequences had a higher increase in HW during post-weaning period compared with lambs of H-L and L-L sequences. Quantity and quality of colostrum was not influenced by pre-pubertal plane of nutrition (P&gt;0.05). Based on the results of current study the authors&rsquo; suggestion is that skeletal compensatory growth during post-weaning period can compensate losses caused by poor nutrition during pre-weaning period. And also, the results indicated that skeletal size is a better indication for first-lactation colostrum yield than body weight (BW). پرونده مقاله

        میزان عامل رشد شبه انسولین(IGF) توسط محرک های مختلف مانند هورمون‌ها، عوامل رشد و شرایط تغذیه‌ای تنظیم می شود. در مطالعه‌ی حاضر، تاثیرسطوح مختلف آنزیم های فیتاز بر بیان ژنIGF-1 در بافت کبدی جوجه های گوشتی مورد بررسی قرار گرفت. آزمایش با 7 تیمار، 5 تکرار و 25 قطعه جوجه در هر تکرار انجام گردید. جیره‌ های آزمایشی شامل جیره شاهد که فسفر مورد نیاز آن توسط دی کلسیم فسفات تامین می‌شد و جیره های حاوی آنزیم های مختلف فیتاز میکروبی تجاری بر پایه ذرت و سویا مورد بررسی قرار گرفت. به منظور بررسی بیان ژن IGF-1 ابتدا کل RNA از بافت کبد استخراج و پس از ساخت cDNA، میزان بیان ژن با استفاده از تکنیک Real time PCR اندازه‌گیری شد. نتایج حاصل از بررسی بیان ژن IGF-I در سطوح مختلف فیتازها اختلاف معنی داری را بین تیمارها نشان داد. بیان ژنIGF-I در تیمار 1و2 شامل فیتاز هوستازیم و فیزایم 50 درصد کمترین میزان بیان و تیمار 6 با فیتازهای هوستازیم و رونوزایم 100 درصد بیشترین میزان بیان را نشان داد. افزایش میزان فیتازها باعث افزایش بیان IGF1-mRNA کبدی شده و می تواند متعاقبا عملکرد صفات رشد را نیز تحت تاثیر قرار دهد. پرونده مقاله
