        پروتئینوگرام‌های سرم 10 گوساله ماده هلشتاین تغذیه شده با شیر از طریق روش‌های متداول (COV) و تغذیه شیر مرحله پایین (step-down) (STP) در سنین مختلف ارزیابی شد. گوساله‌های تغذیه شده با استفاده از روش COV در طول 24 ساعت اول با آغوز تغذیه شدند که آنها با مادرهایشان مانده بود More
        پروتئینوگرام‌های سرم 10 گوساله ماده هلشتاین تغذیه شده با شیر از طریق روش‌های متداول (COV) و تغذیه شیر مرحله پایین (step-down) (STP) در سنین مختلف ارزیابی شد. گوساله‌های تغذیه شده با استفاده از روش COV در طول 24 ساعت اول با آغوز تغذیه شدند که آنها با مادرهایشان مانده بودند و برای دو روز دیگر و بعد جایگزین شیر دریافت کردند، و به مقدار 4 لیتر در روز برای 59 روز شیر دریافت کردند. گوساله‌ای تغذیه شده با استفاده از روش STP با آغوز تغذیه شدند و سپس شیر به شکل زیر: 6 لیتر شیر در هر روز از روز 6 تا 25 بعد از تولد، 4 لیتر شیر در روز از روز 26 تا 45، و 2 لیتر شیر در روز از روز 46 تا 59 بعد از تولد. نمونه‌گیری خون در سنین 7، 21، 28، 42، 49 و 56 روزگی انجام شد. برای گوساله‌های تغذیه شده با استفاده از هر دو روش تغذیه با شیر، غلظت آلبومین سرم افزایش داشت، اگرچه غلظت بتا-گلوبولین سرم با افزایش سن گوساله کاهش داشت. گوساله‌های تغذیه شده با استفاده از روش COV غلظت‌های پروتئین تام سرم، گلوبولین، بتا-گلوبولین، و گاما گلوبولین بیشتری داشتند، اگرچه غلظت‌های A/G، آلفا 1-گلوبولین، و آلفا 2-گلوبولین برای گوساله‌های در گروه STP بالاتر بود. این نتایج برای ایجاد پروفایل پروتئین سرم گوساله‌ها مهم است که به‌ طور معنی‌داری توسط روش‌های تغذیه شیر و سن گوساله تحت تأثیر قرار می‌گیرد.         Background & Objectives: Citrus psorosis virus is the causal agent of one of the most important viral citrus diseases in the world and has been detected from the citrus orchards in the east of Mazandaran province, IRAN. Because of the simultenious presence of various strains of this virus in infected citrus samples, molecular detection methods like PCR are not able to detect this virus with high accuaracy. In this research, we introduce a method for the precise detection of Citrus psorosis virus. Materials & Methods: Detection and Identification of infected samples performed by triple antibody sandwich indirect-ELISA (TAS-ELISA) method and monoclonal antibody from symptomatic Thomson orange and Unshiu tangerine samples with suspicious symptoms to CPsV infection in Sari, Neka and Ghaemshahr districts of Mazandaran. Infection in samples tested by ELISA and confirmed by RT-PCR. Extraction of CPsV-dsRNA (Double-strand RNA) from infected samples was done by CF-11 column. Molecular hybridization performed by DIG labeled cDNA probe. Results: Exploring the electrophoretic pattern of dsRNA extracted from the CPsV infected citrus samples by using ELISA and RT-PCR methods, indicating the presence of CPsV genomes with the molecular weights of 920 bp and 2500 bp in infected samples. The presence of CPsV in the samples was also confirmed by using molecular hybridization and designed probe.   Conclusion: The major advantages of the hybridization method relating to the dsRNA electrophoretic mobility is the simultenious detection of the all citrus psorosis virus strains within the infected citrus samples.         In the present study, the HA-TiO2 nanostructured composite coatings with 0, 10 and 20 wt% TiO2 were fabricated by electrophoretic deposition at 20 V for 3 min. Electrochemical corrosion behavior of the samples was conducted in SBF solution at 37oC by potentiodynamic polarization tests. For the effect of titania on apatite formation, electrochemical impedance spectroscopy after in-vitro tests were conducted on coatings. The variation of electrochemical parameters in equivalent circuit with different times of apatite formation was discussed for each sample. The lowest corrosion current density (icorr) and the highest corrosion potential (Ecorr) and linear polarization resistance (Rp) were obtained for HA-20 wt% TiO2 sample. The EIS analysis revealed that the total resistance of HA-20 wt% TiO2 sample is higher than that of other samples. The ICP analysis of Ca2+ concentration of SBF solution showed that the highest dissolution rate was achieved for HA sample. The concentration of Ca2+ of HA-20 wt% TiO2 sample remained almost constant after 15 days immersion which indicated the accelerated apatite formation. Moreover, the bonding strength of coating was increased by a factor of 2 with the addition of titania in HA-20 wt% TiO2 sample. Manuscript profile

        Single layer and functionally graded coatings of hydroxyapatite and TiO2 particles were deposited on Ti-6Al-4V titanium alloy substrate by electrophoretic deposition technique (EPD). The coatings chemical composition and morphology were investigated using energy dispersive X-ray spectroscopy (EDX), scanning electron microscope (SEM) and X-ray diffraction (XRD) methods. The results showed in HA/TiO2 functionally graded coating, the composition varied from inner layer to top layer as 100 %wt TiO2 to 100 %wt HA. To investigation biocompatibility of the coatings, the culture Wharton’s Mesenchymal Stem Cells was used. The results showed the HA/TiO2 functionally graded coating and HA single layer coating were more biocompatible in comparison to TiO2 single layer coating. The adhesion strengths of the coatings were measured by shear testing and the results showed that the HA/TiO2 functionally graded coating has more adhesion strength (~31 MPa) compare to HA single layer.         The aim of this research is to investigate the stability of SiC nanoparticles for deposition on C/C composite by EPD method. Thus, different suspensions were prepared with different solvents and in presence of PEI as dispersant. The stability of suspensions was studied by using their macroscopic pictures after 24 h, and measurement of zeta potential values and particle size distribution. Thereafter, the coating was applied onto the substrate using the optimized suspension. The results showed that the suspension prepared in ethanol, with pH=10 and addition of 6 wt% PEI has the best stability. Also, SEM micrographs revealed that the coating applied with the voltage of 30 V results in a better quality and has a uniform and pitiless surface. Manuscript profile

        In the present study, hydroxyapatite (HA) Single layer TiO2 single layer, HA-TiO2 double layer and functionally graded HA/TiO2 coatings were deposited on Ti-6Al-4V titanium alloy substrate by electrophoretic deposition technique (EPD). Impact resistance of coatings were measured by ball drop impact test and the results showed that the HA/TiO2 functionally graded coating has more impact resistance compare to HA single layer and HA-TiO2 double layer coatings. The wear resistance of coatings were investigated by pin on disc method. The normal load and sliding distance were 0.5 N and 50 m respectively and the worn surface was observed using scanning electron microscope (SEM) and analyzed by ImageJ software. The results showed that the HA/TiO2 functionally graded coating has more wear resistance compare to HA single layer. Additionally, investigation of worn surface of coatings showed that all of the coatings have abrasive mechanism but wear mechanism for functionally graded coating, changed and delamination was added to abrasive mechanism.         Abstract In this research, Fabrication of YSZ/Al composite coating on Incoloy 825 superalloy using electrophoretic deposition has been investigated. Dispersion of YSZ and Al particles suspension in acetone in presence of iodine, as dispersant, was studied by particle size and zeta potential measurement. According to the results, the suspension containing 1.2 g/l iodine has been chosen for electrophoretic co-deposition of YSZ and Al particles. Mean diameter of YSZ and Al particles in this suspension was measured 111.6 nm and 2.658 μm and zeta potential value of these particles was measured 50.2 and 16 mV, respectively. In order to investigate the influence of applied voltage and deposition time on quality of formed deposit, electrophoretic co-deposition has been carried out at different voltages and deposition times. Results revealed that the deposit formed at voltage of 6 V and deposition time of 3 min had uniform and crack-free surface. SEM image showed that this coating had thickness of 19.63 μm.         The kinetics of electrophoretic deposition is influenced by various factors. Hamakr summarized them on five parameters such as suspension concentration, electric field on the suspension, the surface area of the electrodes, electrophoretic mobility, and the process time. The electric field in the suspension can be changed using the electric potential applied between the electrodes and electrode distance change. Since the suspensions is a non-ohmic resistance, these two parameters will not be the same. The change of electric field using each of these two parameters causes kinetic equations change. In long distances electrodes and poor electrical potential, kinetic equations are based on electrical resistance more accurately. In the strong fields and low electrode distance kinetic equations based on equivalent conductivity are more accurate. The results of this study showed that, in the electric field 70V/cm Ferrari et al. equation and in the electric field 25V/cm Sarkar and Nicholson equation have accuracy of less than 0.01 and less than 0.1, respectively.         In present research, genetic diversity of gliadins among 30 wheat landraces belong to region of Zanjan-Iran, was studied by Acid-PAGE technique. Totally 54 polymorphic gliadin bands were identified. The highest frequency was related to gliadin band 46 witch observed in 90% of wheats. Also gliadins 12.5 and 15.5 were observed in 86.7% of the wheats. Genetic diversity of the wheats based on total gliadin found to be high (H=0.967). Highest and Lowest genetic variation related to omega-gliadins (H=0.917) and gamma-gliadins (H=0.781) respectively. Wheats of Zanjan showed diversity in terms of gliadin bands number (11-25 bands). The entire wheats, showed specific gliadin pattern. Totally, 29 omega-gliadin patterns, 15 gamma-gliadin patterns, 22 beta-gliadin patterns and 23 alpha-gliadin patterns were observed. Cluster analysis could separate three tetraploid genotypes TN-11372, TN-11734, TN-11743 from hexaploids. These results indicate that gliadin proteins analysis is useful for evaluating of genetic diversity, characterizing of wheat genotypes and separating of different ploidy levels. Manuscript profile
