Hormonal, biometric and histological investigation of testis after allograft transplantation in rams

Abstract :
Introduction: Stem cells are undifferentiated cells with ability to renew and divide indefinitely to generate specialized cells. The aim of present study is to hormonal, biometric and histological investigation of testis after allograft transplantation in rams. Materials and Methods: In the present study, 5 male lambs (less than 6 months old) were used as recipients, 5 lambs as donors, 5 lambs as to confirm the effectiveness of tamoxifen and 5 lambs as controls. Azoospermia was induced in five lambs by orally administration of tamoxifen. Adipose tissue from 5 lambs was biopsied and cells were isolated and cultured. This followed by a transplantation of mesenchymal stem cells onto recipients. The Blood sampling and measuring of circumference of scrotum and the dimensions of testis were done three stages before and after the induction of azoospermia and after transplantation. 49 days after transplantation, one testicle was removed for histological evaluation. Results and Discussion: The highest and lowest scrotal circumference sizes were related to control at age of 9 months and treatment group at age of 7 months (p<0.05). Effect of applying factor, sampling time and treatment × time on all studied traits was significant. Induction of azoospermia using tamoxifen led to severe destruction of germinal epithelium in seminiferous tubules. Conclusion: Transplantation of stem cells obtained from adipose tissue can induced spermatogenesis in seminiferous tubules of azoospermic sheep. Therefore, this technique can be used for the transfer of stem cells from a superior animal as a donor for production of functional sperm in another.
