Molecular detection of Cryptococcus neoformanse in pigeon droppings collected from west Tehran city

Abstract :
Cryptococcus neoformans is an encapsulated yeast belongs to basidiomycota that causes a life-threatening disease, cryptococcosis, in immunocompromised hosts. C. neoformans classifies to three varieties which habitat on pigeon droppings and eucalyptus trees. because of it’s role in cryptococcal infections in immunocompromised patients, detection of this funge in natural samples like pigeon droppings has got a great importance. The aim of this study is a molecular detection of C. neoformans in pigeon dropping specimens by standard PCR containing internal control. Diagnosis of C. neoformans in pigeon droppings was done by DNA extraction of specimens via boiling. A part of C. neoformans rDNA along with its specific primers include ITS1 and CN4 – all as a 415bp segment- was used as the target gene in PCR test. PCR test’s specificity and sensitivity were optimized. Then by using a competitive internal control, PCR test was performed on 100 collected pigeon dropping specimens.Out of 100 tested specimens, 11 positive results were come out after PCR test with internal control. Test’s sensitivity for C. neoformans was 10 DNA molecules. No other product was amplified through PCR test which proves test’s 100% specificity.In according to this project’s results, it is culminated that PCR test is a fast, reliable method to detect C. neoformans existence in natural specimens like pigeon droppings
