Cytotoxicity activity of extract of of Viscum album L. on MCF-7(cancer cell line)

Abstract :

Viscum album L. belonging to Loranthaceae family, is a semi-parasitic plant which grows on various host treeand shrubs. There are different reports about various biological activities in V.album such as antidiabetic,antioxidant, vasodilator, sedative, antihypertensive, antiepileptic, antipsychotic, antiinflammatory,immunostimulant, antimutagenic as well as anticancer. The objective of this study was to examine the in vitrocytotoxic activities of a crude ethanol extracts prepared from this plant, on human MCF-7 cell line by Dyeexclusion and Micro culture tetrazolium test (MTT) cytotoxicity assay. The media were replaced with 31.25,62.5, 125, 250, 500, 1000 and 2000 g/ml concentrations of crude extract, no extract was added to the negativecontrol well. The optical density (OD) colored solution was quantified at 570 nm wavelengths by an ELISAReader, after 24 and 48h incubation. Also percent growth inhibition of cells exposed to extractions wascalculated. The results showed that ethanolic extracts, after 24h incubation, at in 250 g/ml and upper doses,suppressed the proliferation of cancerous MCF-7 cells by significant difference with control group (p<0.05).Maximum inhibitory effects were found at 2000 g/ml (34.30%). Also, after 48h incubation, totalconcentrations had significant different with control group in inhibition effect; and maximum inhibitory effectswere showed at 2000 g/ml (99.50%). Results showed the V. album extracts dose dependently suppressed theproliferation of MCF-7 cells. Therefore ethanol extract of V. album able to inhibit the growth of MCF-7 cell lineand cytotoxic effects is very high.
