Application of PCR-RAPD molecular on sex identification in white fish (Rutilus frisii Kutum) from the Caspian Sea

Abstract :
In order to identify the sex marker in white fish, samples from 5 male and female specimens were collected andtheir genomic DNA were determined using spectrophoresis on 1% agaroz gelFollowing Ethidium Bromide staining. Genome structure of White fish was studied by conducting PolymeraseChain Reaction (PCR) using DNA and 124 sets of primers, 10 nucleotides each and PCR products wereevaluated by polyacrylamid gel electrophoresis following silver nitrate staining. All bands were numbered using1 and 0 scores corresponding to having and not having a band respectively and data analyzed by RAPDPLOTprogram.Results of present study indicate that 44 sets of primers have not shown any flanking site and produced no bands,while 80 sets of primers produced sharp and visible bands on polyacrylamid gel. At present study, totally 1600bands were scored. In comparison the band position in all samples, results showed none of bands wascorresponds to the specific male or female fish. According to the results and statistics obtained. It hash may bedue to absences of sex chromosome in this species or genes corresponding to sex differentiation are spread ondifferent autosomal chromosome which commonly with environmental factors contributes to sex determination.
