1 - Application of response surface methodology (RSM) for the optimization of supercritical CO2 extraction of Elaeagnus protein: a comparison study

In this study, dried fruit protein was extracted using supercritical carbon dioxide method. Extraction was performed at different supercritical temperatures, supercritical pressure, and solvent ratio of carbon dioxide and methanol. According to the ANOVA results, the optimum operating condition for the extraction of protein with supercritical CO2 was as following:150.85 bar, 51.06°C, 59.80 min and methanol aqueous solvent concentration of 717.08 µL, leading to extracting of 47.53 mg protein per 10 g sample. Compared to other extraction methods, the amount of extracted protein was high with this method. In addition, less damage was done to other physical-chemical properties of the protein, including protein structure, amino acid chains, etc. Among the operating conditions, supercritical pressure had the highest effect on proteins. Therefore, the results of this study suggested that the supercritical carbon dioxide extraction can be sued as an effective method for protein extraction from plants. تفاصيل المقالة -

2 - بررسی فیتوشیمیایی و آنتیاکسیدانی عصاره میوه گیاه دارویی سنجدL. Elaeagnus angustifolia در رویشگاههای مختلف منطقه شاهرود
اسماعیل باباخانزاده سجیرانی سید جواد موسوی زاده خدیجه مظفریگیاه سنجد (Elaeagnus angustifolia L.) به علت وجود متابولیتهای ثانوی فلاونوئید و فنول از خواص آنتی اکسیدانی برخوردار است و به این جهت دارای عملکردهای دارویی فراوان است. از آنجایی که کمیت و کیفیت آن مواد در شرایط آب و هوایی مختلف متغیر میشود، این تحقیق با هدف بررسی تغیی أکثرگیاه سنجد (Elaeagnus angustifolia L.) به علت وجود متابولیتهای ثانوی فلاونوئید و فنول از خواص آنتی اکسیدانی برخوردار است و به این جهت دارای عملکردهای دارویی فراوان است. از آنجایی که کمیت و کیفیت آن مواد در شرایط آب و هوایی مختلف متغیر میشود، این تحقیق با هدف بررسی تغییرات فیتوشیمیایی و آنتیاکسیدانی عصاره میوههای گیاه سنجد در رویشگاههای مختلف منطقه شاهرود (مجن، میامی و کلامو 1000 - 1400 متر) انجام گرفت. عصاره متانولی میوهها با استفاده از روش خیساندن، میزان فنل و فلاونویید کل در عصارهها با استفاده از روشهای اسپکتروفوتومتری، ارزیابی و مقایسه عملکرد آنتیاکسیدانی با استفاده از روش DPPH انجام گرفت . نتایج نشان داد با افزایش ارتفاع در منطقه کوهستانی مجن (1400 متر)، میزان ترکیبات فنولی، فلاونوییدی و فعالیت آنتیاکسیدانی میوه گیاه افزایش معنیداری پیدا میکند و این موضوع در تایید اثر متقابل تنشهای محیطی موجود در ارتفاعات مختلف رویشگاهی، بر کمیت و کیفیت مواد موثره دارویی گیاهی قابل بحث است. تفاصيل المقالة -

3 - بهینهسازی و مقایسه اثر شرایط مختلف استخراج (فراصوت و خیساندن) بر ترکیبات شیمیایی عصاره میوه گیاه Elaeagnus angustifolia L.
مریم سردرودیان معصومه مهربان سنگ آتش اکرم آریان فرسنجد با نام علمی Elaeagnus angustifolia L..، بهدلیل ترکیبات فلاوونوئیدی، سیتوسترولها، گلیکوزیدهای قلبی و ترپنوئیدها از مهمترین گیاهان دارویی و مغذی محسوب میشود. هدف از این پژوهش، بهینه سازی شرایط استخراج ترکیبات فنولی و فلاونوئیدی از عصاره متانولی میوه کامل سنجد (پوس أکثرسنجد با نام علمی Elaeagnus angustifolia L..، بهدلیل ترکیبات فلاوونوئیدی، سیتوسترولها، گلیکوزیدهای قلبی و ترپنوئیدها از مهمترین گیاهان دارویی و مغذی محسوب میشود. هدف از این پژوهش، بهینه سازی شرایط استخراج ترکیبات فنولی و فلاونوئیدی از عصاره متانولی میوه کامل سنجد (پوست، گوشت و دانه) توسط روش فراصوت و مقایسه آن با روش خیساندن بود. روش سطح پاسخ جهت بهینهسازی شرایط استخراج با هدف بالاترین میزان استخراج ترکیبات فنولی و فلاونوئیدی استفاده شد. آزمونها بر مبنای طرح Box-Behnken با سه متغیر مستقل، شامل نسبت حلال به نمونه (50-20 میلیلیتر حلال بر گرم نمونه)،دما (60-30 درجه سانتیگراد) و زمان استخراج (35-5 دقیقه) طراحی شدند. اندازهگیری محتوای فنل کل عصارهها با استفاده از معرف فولین سیو-کالچو و اندازهگیری محتوای فلاونوئید کل عصارهها بر اساس روش کلریمتری آلومینیوم کلراید انجام شد. ضرایب واریانس بهدست آمده نشان داد اثر دمای فراصوت بر راندمان استخراج نسبت به دیگر عوامل موثرتر بود (05/0p<). شرایط بهینه استخراج بهترتیب: 79/31 (میلیلیتر حلال بر گرم نمونه)، 87/52 (درجه سانتیگراد) و 44/25 (دقیقه) بود. مقدار بازده استخراج، میزان ترکیبات فنولیک و فلاونوئیدها در این شرایط به ترتیب 58/21 (میلیگرم عصاره به ازای گرم نمونه)، 31/106 (میلیگرم اسید گالیک بر گرم ماده خشک) و 98/32 (میلیگرم کوئرستین بر گرم ماده خشک) بود. مقایسه بین دو روش استخراج، نشان داد که بازده استخراج ترکیبات موثره گیاه در روش فراصوت نسبت به روش خیساندن بیشتر است. بنابراین استفاده از روش فراصوت جهت استخراج ترکیبات فنولیک از میوه سنجد پیشنهاد میشود. تفاصيل المقالة -

4 - Investigation of physicochemical compositions of Elaeagnus angustifolia L fruit for using in food industry

Elaeagnus fruit contains a wide range of different food components. Recent researches indicated that there are remedial properties in this plant and, is used as painkiller in patients with rheumatoid arthritis and seizures. With respect to the medicinal and nutritional properties of elaeagnus as an appropriate additive, researches about its application in the food industry is of special importance. In this study, the chemical and rheological properties of sugar variety of elaeagnus for using in food processes were examined.Experimental: For this purpose, the perfect flour of the elaeagnus was evaluated in terms of its chemical and rheological properties in the flour system using Farinograph.Results & Discussion: The results indicated that elaeagnus fruit have crude fiber (4.2%), fat (2.0%), ash (1.9), protein (7.8%) and suger (48.9%). The Farinography test results showed that increasing of stability of wheat flour dough containing elaeagnus flour by 40% in comparison with the control sample. Improving the rheological properties of dough was due to the increased amount of protein, fat, crude fiber and minerals of flour containing 10% elaeagnus flour in comparison with the control sample.Industrial and practical Recommendations: The results of this study indicated that different combinations of elaeagnus fruit for using in various food products and its effect on rheological properties of dough and its maintenance conditions. تفاصيل المقالة -

5 - Essential Oil Composition of Elaeagnus angustifolia and Elaeagnus orientalis from Sistan and Baluchestan, Iran

In folk medicine, the fruit and flower of these plants are used as a tonic and antipyretic agent. The aim of the present study was to chemical analysis of volatile constituents of E. angustifolia and E. orientalis flowers. Experimental: The essential oils of E. angustifolia and E. orientalis flowers collected from Zahedan in Sistan and Baluchestan province were obtained by hydro-distillation. Chemical composition of oils were determined by GC/MS. Results & Discussion: 12 compounds representing 100% were identified in the essential oil of E. angustifolia flowers in which Ethyl cinnamate (73.24%(, Cyclohexanecarboxylic acid, ethenyl ester (6.94%(, Isobutyl cinnamate (4.74%), Palmitic acid (4.39%), Buthyl cinnamate (2.38%), Ethanol (2.24%) and Methyl 9,9- Dideutero- Octadecanoate (2.05%( were the major constituents. In the essential oil of E. orientalis flowers twelve constituents representing 100% were identified in which Ethyl cinnamate (84.80 %(, Ethanol (7.86%), Tetra hydrogeranyl acetone (2.18%) and Phenantrenol (1.26%) were the major ones. The results showed that there are some differences and similarities between the oil compositions of Elaeagnus species growing in the same weather conditions. Although the main component of the essential oils of Elaeagnusspecies was Ethyl cinnamate, but its relative content was higher in the oil of E. orientalis. The percentage of Ethanol in the oil of Elaeagnusspecies also was less than 10%. Moreover, although the Isobutyl cinnamate, Buthyl cinnamate and Methyl 9, 9- Dideutero- Octadecanoate compounds were the main component of the essential oil of E. angustifolia flowers, but these compounds were not identified in the essential oil of E. orientalis. Industrial and practical recommendations: The results of the present study showed that the E. angustifolia and E. orientalis flower oils can be utilized as cheap commercially sources to isolation Ethyl cinnamate. (E. angustifolia L.) whole fruit ethanolic extract was investigated on common pathogenic microorganisms (Staphylococcus aureus, Escherichia coli, Enterococcus faecalis, and Candida albicans). The phytochemical composition of E. angustifolia was screened by Gas Chromatography-Mass Spectrometry (GC/MS). The minimum inhibitory concentration (MIC) and the minimal bactericidal concentration (MBC) of the extract were determined using the broth dilution technique. According to the results, 13 major compounds such as flavonoid, aldehyde, alcoholic and fatty acids were identified by GC/MS. In addition, the extract could inhibit the growth of all examined pathogenic strains. The MIC was 3.75 to 1.87 mg ml-1 while maximum activity was found against Staphylococcus aureus. Besides, the MBC was ranging from 7.5 to 3.75 mg ml-1. Since E. angustifolia whole fruit ethanolic extract contains phytochemical compounds and has antimicrobial potential it can be recommended as a natural active agent for application in the food industry. تفاصيل المقالة
