بررسی استفاده از آب در استخراج سیلیمارین از دانههای گیاه خارمریم (Silybum marianum L. Gaertn)

چکیده مقاله :
سیلی مارین ترکیبی دارویی است که از دانههای گیاه دارویی خار مریم (Silybum marianum (L.) Gaertn) استخراج و در درمان بیماری های کبدی, دیابت نوع П و کلسترول بالا استفاده می شود. به منظور استخراج سیلی مارین از حلال های آلی همچون متانول, اتانول, استونیتریل و استون استفاده میگردد. در این تحقیق به منظور مقایسه استفاده از حلال آب با توجه به ارزانی, سهولت و بی خطر بودن به عنوان یک حلال مناسب پیشنهاد شده است. لذا با روشهای مختلف, مقدار و نوع ترکیبهای فلاونولیگنانی عصاره آبی استخراج شده از دانههای گیاه خار مریم توسط روش کروماتوگرافی مایع با کارایی بالا اندازهگیری شد. نتایج نشان داد که بیشترین مقدار تاکسی فولین, سیلی کریستین و سیلی دیانین استخراج شده (به ترتیب 6/0، 9/0 و 3/2 میلی گرم در گرم ماده خشک) با استفاده از روش سوکسله با آب بعد از انجام یک مرحله چربیزدایی میباشد. بیشترین مقدار سیلی بین و ایزوسیلی بین استخراج شده (به ترتیب 24/0 , 15/0 میلی گرم در گرم ماده خشک) با استفاده از روش سوکسله با آب بدون انجام چربی زدایی قبل از استخراج میباشد. در مقایسه با متانول به عنوان حلال مقدار ماده استخراجی با آب بسیار پایین تر بود. به منظور افزایش بازده, با تغییر روش وافزایش زمان استخراج میتوان در آینده به نتایج بهتری دست یافت.
The seeds extract of Silybum marianum (L.) Gaertn is named silymarin and used in treatment of liver diseases, diabetes (II) and hypercholesterolemia. There are different solvents (methanol, ethanol, acetonitrile and acetone) for extraction of this compound. Water is an inetersting alternative because of its low operating and disposal costs. This study was undertaken to evaluate the efficiency of different methods on silymarin production and followed by HPLC analysis of extracted flavonolignans. The highest content of taxifolin, silychristin and silydianin were 0.63, 0.9 and 2.35 mg g-1 DW, respectively in a soxhlet extraction with water for 12 h after a pre-treatment step involving defatting with petroleum ether. The highest content of silybin and isosilybin were extracted 0.24 and 0.15 mg g-1 DW in a soxhlet extraction with water without defatting process. Although the ratio of the extracted componds with water were less than methanolic extraction, for increasing the quality of the products, the water extraction steps should be well studied both in terms of time and methods.
