ترکیبات اسانس، خاصیت ضدباکتریایی و فعالیت آنتیاکسیدانی اسانس اکوتیپهای مختلف Nigella sativa L. در رویشگاههای مختلف ایران

ملخص المقالة :
سیاه دانه با داشتن ترکیباتی از جمله آلکالوئید، فلانوئید و تانن، یکی از بهترین منابع آنتی اکسیدانهای طبیعی به شمار میرود، به همین منظور این مطاله درجهت بررسی ترکیبات شیمیایی، خاصیت آنتی باکتریال و فعالیت آنتی اکسیدانی توده های مختلف این گیاه در سال 1393 در دانشگاه آزاد شهر کرد اجرا گردید. در این آزمایش تودههای مختلف این گیاه از مناطق مختلف ایران (لردگان، شهرضا، سمیرم، فلاورجان، مشهد، کاشمر و سبزوار) جمع آوری شدند. تهیه اسانس به روش تقطیر با آب و شناسایی ترکیبات شیمیایی اسانس با استفاده از دستگاه (GC/MS) انجام گرفت. ظرفیت آنتی اکسیدانی اسانس با استفاده از روش DPPH ارزیابی و با آنتی اکسیدان BHT مقایسه شد. آزمون ضد باکتریایی به روش رقت سازی و علیه باکتری های گرم مثبت باسیلوس سرئوس، استافیلوکوکوس اورئوس، انتروباکتر آئروژینوسا و باکتری های گرم منفی پرتئوس ولگاریس، پسودوموناس آئروژینوسا، اشرشیاکلی صورت گرفت. نتایج نشان داد، مهمترین ترکیبات تشکیل دهنده ی اسانس شامل آلفا توژن، آلفا پینن، بتا پینن، پاراسیمن، گاما ترپینن، آلفاترپینئول و تیموکینون بوده که دو ترکیب تیموکینون (66/14درصد) و پاراسیمن (37/44 درصد) بیشترین مقدار نشان دادند، که این ترکیبات با توجه به موقعیت جغرافیایی محل رویش مقدار متفاوتی دارند. تودههای مشهد و سبزوار بلاترین و توده کاشمیر کمترین خواص آنتی اکسیدانی را نشان دادند. همچنین اثر ضدباکتریایی قوی اسانس سیاه دانه بر باکتریهای سودوموناس آئروژینوسا، آنتروباکتر آئروژینوسا، پرتئوس ولگاریس و استا فیلوکوکوس ارئوس در مقایسه با عوامل ضد میکروبی بیشتر بوده که این مورد در توده کاشیمر بیشتر ار بقیه تودهها بود.
