چکیده مقاله :
زمینه و هدف: کم کاری تیروئید باعث اختلالات حافظه و یادگیری و کاهش عملکرد شناختی میشود. این مطالعه به منظور تعیین تاثیر دوسین (سیاهدانه و عسل) بر روی یادگیری و حافظه در موش های صحرایی نر بالغ بدنبال القای کم کاری تیروئید انجام شد. مواد و روش ها: در این مطالعه تجربی، 60 سر موش صحرایی نر بالغ از نژاد ویستار با وزن تقريبي g 200-225 و سن حدود 3 -5/2 ماه به شش گروه ده تایی تقسیم شد. گروه کنترل (Ctl) هیچ دارویی دریافت نکرد. گروه شاهد (Sham) دریافت کننده ی یک میلی لیتر آب مقطر به مدت 21 روز ، گروه تجربی 1 كم كاري تيروئيد (Hypo) كه داروي متيمازول به میزان 25 میلی گرم بر کیلوگرم بصورت گاواژ به مدت 10 روز دريافت كرد، گروه تجربی 2 دریافت کننده ی دوسین به میزان 100 میلی گرم بر کیلوگرم به مدت 21 روز ، گروه های تجربي 3 و 4 که علاوه بر القای هیپوتیروئیدیسم به ترتيب 100 و 200 میلیگرم بر کیلوگرم به مدت 21روز بصورت گاواژ دریافت کردند. اختلالات حافظه و یادگیری در حیوانات با آزمون ماز آبی موریس ارزیابی شد. نتایج: در ماز آبی موریس، زمان و مسیر طی شده برای رسیدن به سکو در گروه تجربی 1(Hypo) نسبت به گروه کنترل (Ctl) در روزهای سوم و چهارم بیشتر بود. زمان و مسیر طی شده در گروه نجربی 3 و 4 در روز سوم به طور معنی داری از گروه تجربی 1 کمتر بود. نتایج حافظه فضایی نشان داد که میانگین زمان سپری شده در ربع هدف در گروه تجربی 1 نسبت به گروه کنترل کاهش معنیداری یافته است. همچنین افزایش معنی داری در میانگین زمان و مسافت سپری شده در ربع هدف در گروه تجربی3 نسبت به گروه تجربی 1 مشاهده شد. نتیجه گیری: نتایج نشان می دهد که درمان با عصاره دوسین ممکن است اثرات مضر کم کاری تیروئید را بر یادگیری و حافظه بهبود بخشد.
Background & Aim: Hypothyroidism causes memory and learning disorders and reduced cognitive function. This study was conducted in order to determine the effect of dosin (Negilla sativa and honey) on learning and memory in adult male rats after induction of hypothyroidism. Materials & Methods: In this experimental study, 60 adult male Wistar rats with an approximate weight of 225-200 g and an age of about 2.5-3 months were divided into six groups of ten. The control group (Ctl) did not receive any drug. Sham group receiving one milliliter of distilled water for 21 days, Experimental group 1 of hypothyroidism (Hypo) who received methimazole in the amount of 25 mg/kg by gavage for 10 days. Experimental group 2 receiving Dosin at the rate of 100 mg/kg for 21 days, experimental groups 3 and 4, in addition to inducing hypothyroidism, received 100 and 200 mg/kg respectively for 21 days by gavage. Memory and learning disorders in animals were evaluated by the Morris water maze test. Results: : In the Morris water maze, the time and distance traveled to reach the platform in the experimental group 1 (Hypo) was higher than the control group (Ctl) on the third and fourth days. The time and the route traveled in experimental groups 3 and 4 on the third day were significantly less than the experimental group 1. Spatial memory results showed that the average time spent in the target quadrant in experimental group 1 decreased significantly compared to the control group. Also, the average time and distance spent in the target quadrant in experimental group 3 showed a significant increase compared to experimental group 1. Conclusion: The results suggest that treatment with dossin extract may ameliorate the deleterious effects of hypothyroidism on learning and memory.
