بررسی فراوانی استافیلوکوکوس اورئوس و میزان شیوع مقاومت به متی سیلین در بین جدایه های این باکتری از شیر گاوهای مبتلا به ورم پستان درسطح استان خوزستان

چکیده مقاله :
استافیلوکوکوس اورئوس (S. aureus) یکی از علل های اصلی ورم پستان در گاوهای شیری و یک پاتوژن جدی است که بر سلامت انسان تأثیر می گذارد. بروز مقاومت به متی سیلین دراین باکتری از نگرانی های مهم و عمده در دامپزشکی و بهداشت عمومی می باشد. هدف مطالعه حاضر فراوانی میزان استافیلوکوکوس اورئوس در نمونههای شیر جمعآوریشده از گاوهای مبتلا به ورم پستان بالینی و تحت بالینی استان خوزستان و همچنین، فراوانی شیوع ژن mecA در بین این جدایه ها بود . در این مطالعه از 300 نمونه شیر مشکوک به ورم پستان از گاوداری های استان خوزستان تنها 31 (33/10 درصد) جدایه هابا استفاده از مورفولوژی
،آزمایش های بیوشیمیایی و با استفاده از ژن اختصاصی ترمونوکلئاز (nuc) به عنوان استافیلو کوکوس اورئوس تایید شد. بررسی فنوتیپی و حضور ژن مقاومت به متی سیلین در جدایه ها با ردیابی mecA و همچنین الگوی حساسیت یا مقاومت آنتی بیوتیکی جدایه ها با روش انتشار از دیسک با استفاده از دیسک های آنتی بیوتیکی پنی سیلین، کلیندامایسین، داکسی سیلین ، سفوکسیتین و تری متوپریم- سولفامتوکسازول انجام شد.. از 31 جدایه استافیلوکوکوس اورئوس تنها 2 (45/6درصد) جدایه واجد ژن mecA بودند. در حالیکه در روش فنوتیپی به عنوان استافیلوکوکوس اورئوس مقاوم به متی سیلین نبودند. نتایج آنتی بیوگرام جدایه ها نشان داد بیشترین مقاومت مربوط به آنتی بیوتیک های تری متوپریم- سولفامتوکسازول و داکسی سیلین ( 45/6 درصد) و کمترین مقاومت نسبت آنتی بیوتیک پنی سیلین (صفر درصد) می باشد.

Staphylococcus aureus (S. aureus) is one of the main causes of mastitis in dairy cows and a serious pathogen that affects human health. Methicillin resistance in this bacterium is one of the major concerns in veterinary medicine and public health. The aim of the current study was the prevalence of Staphylococcus aureus in milk samples collected from cows with clinical and subclinical mastitis in Khuzestan province, as well as the prevalence of the mecA gene among these isolates. In this study, out of 300 milk samples suspected of mastitis from cattle farms in Khuzestan province, only 31 (10.33%) isolates using morphologyIt was confirmed as Staphylococcus aureus by biochemical tests and using specific thermonuclease (nuc) gene.. Phenotypic investigation and the presence of methicillin resistance gene in isolates by tracking mecA, and also the pattern of antibiotic sensitivity or resistance of isolates by diffusion method. From the disc, it was done using penicillin, clindamycin, doxycillin, cefoxitin and trimethoprim-sulfamethoxazole antibiotic discs. out of 31 Staphylococcus aureus isolates, only 2 isolates (6.45%) had the mecA gene. While in the phenotypic method, they were not resistant to methicillin as Staphylococcus aureus. The results of the antibiogram of the isolates showed that the highest resistance was related to the antibiotics trimethoprim-sulfamethoxazole and doxycillin (6.45%) and the lowest resistance was penicillin (0%).
