Citrus genus is known as one of the world's major economic agricultural products. In the current research, a method is reported for artificial seed production technology by shoot tips in Citrus sinensis.var. Thamson navel. Alginate is one of the most efficient gelling agents in the production of capsule coating in artificial seeding technique. In the current research, impact of different constituents of alginate matrix was investigated on artificial seed germination. In order to produce artificial seeds, shoot tips of Thamson navel were coated by three alginate coating including SA1 containing liquid MS and BA and NAA hormones, SA2 with liquid MS without hormone, and SA3 with distilled water. Alginate coating with BA and NAA hormones was used as control environment. The artificial seeds were then stored in the refrigerator (4 ). Following passing storage period, seeds were placed in MS solid medium with BAP (10 mgl-1) and NAA (1 mgl-1) for conversion to seedlings. Using distilled water instead of MS medium for the preparation of alginate coating significantly reduced the percentage of conversion into seedlings. پرونده مقاله

The synthetic seed technology is becoming popular due to its wide application in germplasm conservation and for exchanges between countries in the floriculture trade. In this study, this method was used to study the germination and conversion capabilities of orchid species Dendrobium White Fairy when stored at different storage times and with different storage containers. A high germination percentage of 80% was observed for encapsulated synthetic seeds up to 150 days of storage in Petri dishes and screw capped polypropylene tubes and this percentage only began to decline gradually from 150 storage days and more. Besides that, synthetic seeds stored in polypropylene tubes were shown to germinate faster and develop into plantlets with longer shoots and roots as compared to those stored in the Petri dishes. This proved the efficiency and suitability of polypropylene tubes over Petri dishes as storage containers. پرونده مقاله

The present study describes the encapsulation of protocorm of Cymbidium bicolor Lindl. from 60 days-old in seed cultures for short- term conservation and propagation. Various concentrations and combinations of gelling matrix (sodium alginate) and complexing agents (calcium chloride) were tested to prepare uniform beads. The ideal beads were ob- tained through a combination of 3% sodium alginate and 100 mM calcium chloride. Encapsulated protocorms exhibited the best re-growth and con- version frequency on MS medium supplemented with BA (4.42 μM). En- capsulated protocorms stored at 25°C were green and retained the viability with potential for conversion (52%) and germination even after 360 days. The encapsulated protocorms stored at 4°C remained viable up to 30 days beyond which the conversion rate decreased drastically. Well-de veloped plantlets were transplanted into plastic pots containing vermiculite and maintained for 60 days in the culture room for acclimatization. The 90% of the recovered plantlets were hardened off and established suc-cessfully in the soil. The present study could be useful for large scale propagation as well as short term storage of this commercial orchid پرونده مقاله
