        1 - تغییرات شاخص‌های جوانه‌زنی و فعالیت سیستم آنتی‌اکسیدانتی بذرهای پنبه (Gossypium hirsutum L.) طی زوال

        بذرها در طی انبارداری زوال یافته و پیر می شوند به طوری که سرعت این فرآیند به دما و رطوبت بذر در طی انبارداری بستگی دارد. این آزمایش به منظور مطالعه فعالیت سیستم آنتی اکسیدانتی در بذرهای پنبه رقم ارمغان در طی زوال صورت گرفت. برای ایجاد سطوح مختلف زوال از آزمون تسریع پیری استفاده شد. بذرهای پنبه به‌مدت 0، 48، 96، 144 و192 ساعت در دمای 43 درجه سانتی گراد و رطوبت نسبی 100 درصد قرار گرفتند. نتایج نشان داد با افزایش دوره زوال هدایت الکتریکی غشاء، پراکسیداسیون لیپید و پراکسید هیدروژن افزایش یافت. افزایش پراکسید هیدروژن همراه با کاهش فعالیت آنزیم کاتالاز و افزایش فعالیت آنزیم پراکسیداز و مقدار اسید آسکوربیک بود که نشاندهنده تاثیر کاهنده دوره زوال بر فعالیت آنزیم کاتالاز نسبت به آنزیم پراکسیداز و اسید آسکوربیک بود. همچنین با افزایش دوره زوال درصد، سرعت و یکنواختی جوانه زنی کاهش یافت. به طور کلی نتایج این تحقیق بیانگر این مطلب است که یکی از دلایل اصلی زوال بذر در بذرهای پنبه، تنش اکسیداتیو ناشی از تجمع گونه های اکسیژن فعال و کاهش کارایی سیستم آنتی اکسیدانتی می باشد. پرونده مقاله

        One of the methods for achieving more yields is to use seed priming technique correctly. When it comes to using seed priming, practically important issues should be taken into account including interval between priming to planting and conditions for storage of primed seeds. Thus, it is very crucial to be informed of duration and temperature for storage of primed seeds. This research seeks to compare the effect of temperature and duration of hydroprimed wheat seeds (var Kouhdasht) on growth indices and grain yield. This test was carried out with three replications in Research Farm of Agricultural Faculty of Lorestan University, as completely randomized block design. The relevant factors include storage temperature (15, 20 and 25° C) and storage duration of hydroprimed seed (0, 2, 4, 6, and 8 days). The results showed that hydropriming increased number of productive spike and grain yield. Nevertheless, delay (8 days storage) in planting and storage seeds at high temperature (25° C) could decrease benefits of priming. Storage temperature and duration of hydroprimed seeds affect morpho-physiological characteristics as well as yield. Seeds stored at 15 and 20° C showed a better yield than those stored at 25° C. Primed seeds stored at 20° C for 2 days showed a better performance than the other applied treatments. پرونده مقاله

        Annual Sainfoin (Onobrychis crista-galli (L.) Lam.) is widely adapted to moderate and cold regions of Iran and naturally grows in pasture and rangelands used for forage in these areas. In order to study the effects of priming on seed germination and seedling growth in O. crista-galli, two factorial experiments were conducted based on a randomized complete design with three replications under laboratory and greenhouse conditions in Research Institute of Forests and Rangelands, Tehran, Iran in 2014-2015. Experimental factors were (A) five conservation methods including medium-term storage (active cold room 4°C for 15 years), long-term storage (basic cold room -18°C for 15 years), regenerated seeds (control) and deteriorated seeds using accelerated ageing techniques )40°C, 98% of RH for 48 and 72h). Levels of factor B were four priming treatments including Control (without priming), two osmopriming (PEG -0.4Mpa and -0.8Mpa), and hydropriming (imbibition with distilled water). Data were collected for germination percent, rate of germination, root length, shoot length, seedling length, vigor index, seedling weight and root/shoot length ratio (R/S). Result of laboratory showed higher mean values of most traits except root length in base store (-18°C). In both experiments, the highest root length was obtained in aged seeds. In greenhouse, higher values of many traits were obtained in osmopriming (PEG -0.4Mpa). In both experiment, higher mean values of many traits were obtained using hydropriming in seeds conserved in both base and active store. In the latter stores, highest root length, seedling length, R/S ratios were obtained by osmopriming (PEG -0.4Mpa and -0.8Mpa). To accelerate aging test, higher mean values of all the traits were obtained by osmopriming (PEG -0.4Mpa). It was concluded that osmopriming could be used as an effective method for the recovery of natural and artificial deteriorated seeds. پرونده مقاله

        Secale montanum is one of the important perennial grasses growingnaturally in arid to semiarid pastures and rangelands, with a typical Mediterraneanclimate, in northern and western Iran at altitudes of 800-2900 m. In this paper, seedstorage protein profiles of nine wild populations of S. montanum from differentregions of Iran and their phenotypically superior progenies as well as a multi-originpolycross (PLC) were studied. High levels of polymorphism were observed over allpopulations with the average number of bands and average heterozygosity. Superiorprogeny of different populations showed less genetic variability than wild parents interms of band diversity, whereas PLC samples showed extremely high values ofgenetic parameters. Two locally common bands were observed in almost all wildparent populations, which are missing in superior progeny of different populationsand PLC. These results provide highly support for the hypothesis that neutral geneticdiversity has been reduced or inadvertently lost via artificial selection. Among wildparent populations and their superior progenies significant differences were observedin expected heterozygosity suggesting that more intensive breeding practices mayhave resulted in a further erosion of genetic variability. Neighbor-joining clusteranalysis showed that wild populations and the phenotypically superior progeny ofdifferent populations were separated into two groups. This suggests that foundereffects and subsequent selection have had more effect on the genetic differentiationbetween these accessions than geographical separation. This technology, seed storageprotein profiling, has great potential for use in breeding programmes. پرونده مقاله

        5 - Study of the Diversity in Different Cultivars of Pistacia vera L. Resistant to Drought and Salinity: Comparing Protein Patterns Using SDS-PAGE Method

        Pistachio is one of the most important agricultural products that have always been associated with Iran, and its production has a long historical background in our country. In this research, protein patterns of 10 cultivars of Pistacia vera L. were compared in which cultivars grown in normal conditions where compared with cultivars grown in salinity and water shortage to determine diversity. For this purpose, after extraction of storage proteins of seeds, the density of proteins was determined using the Bradford method. Then, the SDS-PAGE method was used to separate extracted proteins. In order to analyze electrophoretic data, we allocated 0 to absence and 1 to presence of each of bands. The results showed that stresses resulted in expression or loss of expression of some of the bands in cultivars, and the total distance between bands in each of the cultivars from origin of bands’ movements from the gels was considered as one of the studied characteristics. The data was analyzed using cluster and Principal component analysis using SPSS software. Analysis of variance showed that average protein concentration of species was 19.59% and that the maximum protein percentage belongs to the Badami cultivar of Rafsanjan with 22.09%, and that the minimum protein percentage belongs to the Fandoghi cultivar of Rafsanjan with 13.98%. There was a significant difference among the amounts of proteins at the 5% level. In qualitative analysis of storage proteins of seeds, a total of 18 bands were observed. The maximum number of protein bands belongs to Kalehghochi and Fandoghi cultivars of Sirjan (saline area). In cluster analysis, species were classified in 10 different groups. پرونده مقاله
