Characterization of phytoconstituents in the 70% methanolic extract of Cissus rotundifolia (Forssk.) Vahl. was performed using high-performance liquid chromatography coupled with electrospray ionization mass spectrometry. Furthermore, tandem mass spectrometry (MS/MS) was performed to assist in the structural elucidation of compounds. Twenty-seven compounds were identified. The identified compounds were four acids, sixteen phenolics, two steroidal saponins, two coumarins, two stilbenoids and one triterpene. Astragalin 1, β-amyrin 2 and 1β-hydroxy-kryptogenin-1-O-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranoside 3 were isolated by column chromatography and their structures were identified by spectral analysis including 1D and 2D NMR. The plant extract and the isolated compounds 1-3 were evaluated for their radical scavenging activity. Compounds 1 and 2 showed significant activity relative to that of ascorbic acid as a standard. The cytotoxicity of the plant extract, the isolated compounds and the standard doxorubicin was evaluated using the three human tumor cell lines HT116, MCF-7 and PC-3. The plant extract exhibited significant cytotoxicity against MCF-7 while showing no activity against the other cell lines. The isolated compounds 1-3 showed no activity against the used cell lines. پرونده مقاله

The aerial parts of Ferula ovina (Boiss.) Boiss. were extracted by methanol and then fractionated with solvents with different polarity. The evaluation of the bioactivity of the plant extracts including antioxidant, antimicrobial and antiproliferative activity was carried out using various assays. The phenolic and flavonoid compound profile of the extract were established using HPLC analysis. The leaves showed the highest total phenolic content and radical scavenging activity; the flowers had the maximum amount of total flavonoid content, and the stems exhibited the maximum ferrous ion chelating ability. Among ten microorganisms, the plant extract prevented the growth of three strains, namely Pseudomonas aeruginosa, Acinetobacter baumannii, and Streptococcus pneumonia. The flower of F. ovina (Boiss.) Boiss shows the best anti-human cervical carcinoma property against HeLa cell lines. Gallic acid was found as the major phenolic compound in the plant extract according to the HPLC analysis. پرونده مقاله

The search for naturally occurring plant-based nutraceutical antioxidants is imperative in the management of arrays of diseases. Herein, the methods involved profiling antioxidant activities and nutritional composition of the methanolic extracts from the leaves of Ficus thonningii Blume (FTH), Jatropha tanjorensis J.L Ellis and Saroja (JTR) and Justicia carnea Lindley (JCN) using spectrophotometric techniques. In this sense, J. tanjorensis J.L Ellis and Saroja extract showed the highest total antioxidant capacity and strong DPPH radical scavenging activity with an IC50 of 10.490 ± 0.0320 μg.mL-1 when compared to ascorbic acid (IC50 of 114.140 ± 5.135 μg.mL-1), while for the hydroxyl radical scavenging activity, F. thonningii Blume and J. carnea Lindl. extracts resembled IC50 values of 523.095 ± 13.629 and 503.733 ± 7.562 μg.mL-1, respectively. A positive correlation was observed between the extracts and ascorbic acid (p < 0.001), as well. In addition, nutrients and minerals were present in appreciable quantities. The obtained results validate the assayed plants extracts as potential radical scavengers and promising nutraceutical antioxidants in ethnomedicine. پرونده مقاله

The role of natural antioxidant compounds in preserving the frying stability of oils is increasingly recognized. Orange (Citrus x sinensis) and lemon peels (Citrus × aurantiifolia) are rich sources of antioxidant components. The objective of the current study was to determine antioxidant properties of orange and lemon peels and evaluate their efficacy in preventing lipid peroxidation in heated and stored unrefined sunflower (Helianthus annuus) and groundnut oils (Arachis hypogaea). Peel powder was added at 0.5 and 1.0% level. The oils were subjected to thermal treatment, stored and analyzed on 0, 10, 20, and 30 days for the determination of free fatty acid (FFA) and peroxide value (PV). The obtained results showed that orange peel had higher total polyphenolic, tannins, β-carotene and total carotene contents. The highest DPPH radical scavenging activity was observed in pure methanolic extract (70%). FFA ranged from 0.03-0.1 and 0.04-0.1% in sunflower oil heated at 60 ºC with 0.5 and 1.0 g peel powder, respectively. FFA was in the range of 0.35-0.45% for groundnut oil samples heated at 60 ºC with 0.5 g peel powder, whereas PV ranged between 3.65 and 8.4 meq/kg. The reduction in lipid peroxidation during storage of heat treated peel powder incorporated samples was remarkable for those heated at 60 °C and stored for 10 days. Lemon peels were also rich in total phenols (716 mg/100 g) and flavonoids (168 mg/100 g) and showed considerable antioxidant activity. At the end of the storage duration, both thermally treated groundnut and sunflower oils showed lesser FFA over the ranges 0.12-0.15% and 0.38-0.52%, respectively and PV ranging from 8.96 to 11.5 meq/kg and from 7.9 to 10.9 meq/kg, respectively than control oils (FFA-0.18-0.9% and PV-12.2-21.2 meq/kg). This study showed that orange and lemon peels may be used as natural antioxidant agents to prevent lipid peroxidation. پرونده مقاله

Research works proposed that radical scavenging activity of flavonoids is due to ring B, andthe remaining part of the molecule can be disregarded. Thus the objective of this work is toobserve whether hydroxytoluenes account the radical scavenging activity of flavonoid and toestablish structural requirements for their activity (as they showed appreciable activity) andelucidate a comprehensive mechanism that can explain their activity and termination. Thus, theradical-scavenging activity of nine hydroxytoluene derivatives against 2,2-diphenyl-1-picrylhydrazyl, DPPH was determined. The relative change in energy (ΔHf) associated with theformation of phenolic radicals and the spin distributions in these radicals were determined usingcomputational programs (Density function theory and Hartree Fock). By correlatingexperimental data with ΔHf, the most active compounds and structural features that areresponsible for their activities were identified. Reaction product of 4-methyl catechol with 2,2-diphenyl-1-picrylhydrazyl, DPPH was isolated and characterized in order to unravel themechanism of termination of most active hydroxytoluenes. Termination enthalpy (ΔH2) ofmethyl-catechols and methyl-hydroquinone, once the termination mechanism explained, wascalculated to understand its role in the radical scavenging activity. پرونده مقاله
