بررسی تأثیر حلالهای مختلف بر میزان فنل و فلاونوئید کل و فعالیت آنتیاکسیدانی اندامهای مختلف گیاه Momordica charantia L. در منطقه سیستان

آنتیاکسیدانها ترکیبهایی هستند که از بدن در برابر آسیبهای ناشی از فعالیت رادیکالهای آزاد محافظت می کنند. ترکیبهای فنلی متابولیت های ثانوی گیاهی هستند که توان آنتیاکسیدانی بالایی دارند. کارلا با نام علمی Momordica charantia L. متعلق به تیره کدوئیان ((Cucurbitaceae، دارای ترکیبهای فنلی است که بهعنوان آنتیاکسیدان عمل می کنند. پژوهش حاضر به منظور بررسی اثر حلالهای مختلف (متانول 70 درصد، استون، اتیل استات، کلروفرم و هگزان)بر محتوای فنل و فلاونوئید کل و فعالیت آنتیاکسیدانی اندامهای مختلف گیاه کارلا در منطقه سیستان در سال 95 انجام شد. عصارهگیری از اندامها با استفاده از حلالهای مختلف و به روش خیساندن انجام گرفت. میزان فنل و فلاونویید کل بهترتیب با استفاده از روشهای فولین - سیوکالتو و روش رنگ سنجی آلومینیوم کلرید اندازهگیری گردید. فعالیت آنتیاکسیدانی عصارهها با استفاده از روشهای دی فنیل پیکریل هیدازیل ((DPPH، قدرت آنتیاکسیدانی احیای آهن (FRAP) و پراکسید هیدروژن (H2O2) اندازهگیری شد. بیشترین میزان فنل در عصاره متانولی میوه و برگ (بهترتیب 33/1±96/36 و 03/3±01/33 میلی گرم گالیک اسید به وزن خشک) و کمترین میزان در عصاره های کلروفرمی، اتیل استاتی و هگزانی به میزان کمتر از 1 میلی گرم بر وزن خشک گزارش گردید. نتایج نشان داد بیشترین میزان محتوای فلاونوئیدی مربوط به عصاره استونی و متانولی برگ (بهترتیب 97/1±95/10 و 2/2±03/10 میلیگرم کوئرستین به گرم وزن خشک) بود. عصاره متانولی میوه در هر سه روش، فعالیت آنتیاکسیدانی بالاتری نسبت به سایر عصارهها نشان داد. بر اساس نتایج این تحقیق، میوه کارلا بهعنوان یک منبع مفید از آنتیاکسیدانهای طبیعی پیشنهاد میشود.
Antioxidants metabolites which are protect the body against damage caused by free radical activity. Phenolic compounds are secondary metabolites in medicinal plants, which have a high antioxidant power. Karela (Momordica charantia L.) is an herb belonging to Cucurbitaceae family, consisted of phenolic compounds which can act as antioxidant. The present study was conducted to investigate the effect of different solvents (methanol, acetone, ethyl acetate, hexane and chloroform) on total phenolic, flavonoids content and antioxidant activities of Karela in Sistan region in 2016. Different extracts of plant parts ( Root, leaves and fruit) were obtained by different solvents by maceration method. Total phenolic and flavonoid contents were measured by Folin–Ciocalteu and Aluminum chloride methods, respectively. Antioxidant activity of extracts were determined by using Diphenyl Picryl hydrazyl (DPPH), Hydrogen peroxide (H2O2) and Ferric Reducing Antioxidant Power (FRAP) assays. The results were showed that the highest phenolic content was observed in fruit and leaf methanol extract (36.96±1.33 and 33.01±3.03 mg gallic acid/g DW respectively) and the least amount of phenol was extracted by ethyl acetate, hexane and chloroform which was less than 1 mg/g DW. The highest flavonoid content was observed in leaf acetone and methanol extract (10.95±1/97 and 10.03±2.2 mg quercitin/g DW respectively). Fruit methanol extract showed higher antioxidant activity than other extracts in three methods. Based on results of this research, Karela fruit can be suggested as useful source of natural antioxidants
