چکیده مقاله :
هدف: این مطالعه با هدف تعیین اثر 12 هفته تمرین HIIT بر بیان ژن GLUT4 در عضله دوقلو، گلوکز و مقاومت انسولین در رت های دیابتی نوع 2 انجام گرفت.
روش کار: جامعه آماری را همه رت های نر ویستار انستیتو پاستور ایران تشکیل می دهند که از بین آنها 14 رت 10 هفته ای در دامنه وزن 20 ± 220 گرم انتخاب شدند و توسط تزریق درون صفاقی نیکوتین آمید + STZ دیابتی نوع 2 شدند. سپس به شیوه تصادفی به گروه های HIIT (12 هفته، 5 جلسه در هفته، 7 = n) و کنترل (7 = n) تقسیم شدند. سطوح ناشتایی گلوکز، انسولین، مقاومت انسولین و بیان ژن GLUT4 در عضله دوقلو دو گروه بعد از آخرین جلسه تمرینی اندازه گیری و توسط آزمون تی مستقل بین دو گروه مقایسه شدند.
نتایج: تمرین HIIT به کاهش معنی دار گلوکز ناشتا و افزایش بیان GLUT4 در عضله دوقلو در مقایسه با گروه کنترل منجر شد. در مقایسه با گروه کنترل، شاخص مقاومت انسولین تغییر معنی داری پیدا نکرد.
نتیجه گیری: اجرای تمرینات HIIT مستقل از مقاومت انسولین، سطوح گلوکز رت های دیابتی نوع 2 را بواسطه افزایش بیان GLUT4 در عضلات اسکلتی متاثر می کند. با این وجود، اجرای مطالعات بیشتر با هدف شناخت دیگر مکانیسم های عهده دار هموستاز گلوکز پیشنهاد می شود.
Objective: This study aimed to determine the effect of high intensity interval training (HIIT) on GLUT4 gene expression in Gastrocnemius muscle, glucose and insulin resistance in type 2 diabetes rats.
Methods: The statistical population consisted of all male Wistar rats of the pastor inistitue of Iran, among which 14 were male 10 weeks old weighing 220 ± 20 g. Type 2 diabetes induced by Intraperitoneal injection of nocotinamid + STZ. Then rats divided randomly into interval (5 times/week/6-week, n=7) and control (n=7) groups. Fasting glucose, serum insulin, insulin resistance and GLUT4 gene expression in Gastrocnemius muscle of both groups were measured after lasted exercise and compared between 2 groups by independent T test.
Results: HIIT decreased fasting glucose and increased GLUT4 gene expression compared with control subjects. Compared to control group, insulin resistance expression did not significant change.
Conclusion: HIIT training independent of insulin resistance affects glucose levels in type 2 diabetic rats by increasing GLUT4 expression in skeletal muscle. However, further studies are suggested to identify other mechanisms responsible for glucose homeostasis.
