Effect of Adding Coenzyme Q10 and Ellagic Acid during Cryopreservation on Post-Thaw Quality of Ram Semen

The aim of this study was to determine the effects of co-Q10 and ellagic acid on sperm parameters after the freeze–thawing of Ghezel ram sperm. Twenty ejaculates from five Ghezel rams were collected in this study. Semen samples, which were diluted with a soybean lecithin (SL) based extender containing 0.25 mM ellagic acid, 0.5 μM co-Q10, 0.25 mM ellagic acid + 0.5 μM co-Q10 and no antioxidant (control), were cooled to 4˚C, frozen in 0.25 mL French straws and stored in liquid nitrogen. Sperm motility characteristics, membrane integrity, abnormal morphology, lipid peroxidation and antioxidant activities (glutathione peroxidase, superoxide dismutase and total antioxidant capacity) were evaluated following freeze-thawing. The results showed that 0.5 μM Co-Q10, improved viability, total motility parameters and decreased abnormal sperm and improved linearity (LIN), curvilinear velocity (VCL), straight-line velocity (VSL) and path velocity (VAP) parameters (P<0.05). Ellagic acid and the treatment with a combination (0.25 mM ellagic acid+0.5 μM co-Q10) improved viability and total motility parameters (P<0.05). The additives did not affect the maintenance of superoxide dismutase (SOD) and glutathione peroxidase (GPx), when compared to the control. It can be concluded that addition of 0.5 μM co-Q10 improved the post-thawing quality of ram semen.
هدف این مطالعه تعیین اثرات کوآنزیم Q10 و اسید الایژیک بر روی پارامترهای اسپرم قوچ قزل بعد از فرآیند انجماد-یخگشایی بود. در این مطالعه 20 دفعه نمونه منی از 5 رأس قوچ قزل اخذ گردید. برای نمونههای منی رقیق شده با لستین سویا مقادیر 25/0 میلیمولار از الایژیک اسید، 5/0 میکرومولار کوآنزیم کیوتن، 25/0 میلیمولار از الایژیک اسید + 5/0 میکرومولار کوآنزیم کیوتن و یک نمونه بدون اکسیدان (کنترل) اضافه شده، پس از خنک شدن تا 4 درجه سانتیگراد، بداخل پایوتهای فرانسوی 25/0 میلیلیتری کشیده شده و پس از انجماد در ازت مایع نگهداری شدند. پس از یخگشایی نمونههای منی، ویژگیهای حرکتی، یکپارچگی غشاء، ناهنجاریهای مورفولوژیکی اسپرمها، پراکسیداسیون لیپیدی و فعالیتهای آنتی اکسیدانی (گلوتاتیون پراکسیداز، سوپراکسید دیسموتاز و ظرفیت آنتی اکسیدانی تام) نمونههای منی ارزیابی شدند. نتایج حاکی از آن بود که در نمونههای دریافت کننده 5/0 میکرومولار کوآنزیم کیوتن پارامترهای زندهمانی و تحرک کل بهبود یافته، میزان اسپرمهای غیر طبیعی کاهش یافته و نیز پارامترهای خطی بودن جنبایی (LIN)، میانگین سرعت در مسیر منحنی (VCL)، سرعت در مسیر مستقیم (VSL) و میانگین سرعت در مسیر (VAP) بهبود یافتند (05/0>P). در نمونههای دریافتکننده الایژیک اسید و یا ترکیبی از الایژیک اسید و کوآنریم Q10 (25/0 میلیمولار الایژیک اسید+5/0 میکرومولار کوآنزیم Q10)، پارامترهای زندهمانی و تحرک کل بهبود پیدا کردند (05/0>P). میزان سوپراکسید دیسموتاز و گلوتاتیون پراکسیداز تفاوت معنیداری بین نمونههای دریافتکننده دو آنتی اکسیدان نسبت به گروه شاهد نداشت. میتوان نتیجه گرفت که افزودن 5/0 میکرومولار از کوآنزیم Q10 باعث بهبود کیفیت منی قوچ پس از فرآیند انجماد یخگشای شد.
