A Study on the Effects of Chilling Period on Sperm Quality of Sannan and Jamunapari Goats

The objective of the present study was to examine the effects of chilling period on semen quality of Sannan and Jamunapari. Semen of goat breeds were collected with artificial vagina and chilled at 5-9˚C. Volume, colour and pH were determined on fresh semen. After dilution, microscopic examination was performed for the progressive motility and sperm viability. Diluted semen of both breed was compared by microscopic examination from day 0 to 3 during chilling. Hemocytometer was loaded with 10 µL of semen to evaluate the sperm count. Eosin stain (1%) was used to assess the viable sperm. Although viability and sperm velocity of both goat breed decreased(P<0.05) in a time dependent manner during chilling, the viability between the Sannan and Jamunapari did not differ significantly. However there was a significant difference for means of sperm velocity (P<0.05). Progressive motility and viability decreased gradually during chilling at 5-9˚C.In order to obtain higher successful rate of insemination, chilled goat semen should be supplied to the farmers within a day.
هدف از مطالعه حاضر آزمودن اثرات دوره سرمایی بر کیفیت منی بزهای سانن و جاموناپاری بود. منی نژادهای بز با استفاده از واژن مصنوعی جمعآوری و در دمای 5 تا 9 درجه سانتیگراد سرد شد. حجم، رنگ و pH منی تازه تعیین شد. پس از رقیقسازی، آزمایش میکروسکپی از نظر تحرک پیشرونده و زندهمانی اسپرم صورت گرفت. منی رقیق شده دو نژاد به وسیله آزمایش میکروسکپی از روز 0 تا 3 سرمادهی مورد مقایسه قرار گرفت. هموسایتومتر جهت ارزیابی شمار اسپرمها با استفاده از 10 میکرولیتر منی بارگذاری شد. از رنگآمیزی ائوزین (1 درصد) برای تخمین اسپرمهای زنده استفاده شد. گرچه زندهمانی و سرعت حرکت اسپرمهای دو نژاد بز به طور وابسته به زمان در طی سرمادهی کاهش یافت (05/0>P)، زندهمانی بین بزهای سانن و جاموناپاری به طور معنیدار تفاوت نداشت. در هرحال، تفاوت معنیداری برای میانگینهای سرعت حرکت اسپرم مشاهده شد (05/0>P). تحرک پیشرونده و زندهمانی به طور تدریجی در طی سرمادهی در 5 تا 9 درجه سانتیگراد کاهش داشت. به منظور کسب نرخ موفق بالاتر تلقیح، باید منی سرمادهی شده بز برای دامداران در طی یک روز فراهم شود.
