        Glioblastoma multiforme is one of the most malignant and common brain tumors, and after current combination therapy, patient survival is relatively poor and the disease is always fatal. This study was aimed at investigating the expression of SCAMP5 as a possible biomarker in GBM and evaluating the target miR of this gene. 50 tissue samples with GBM form before treatment and 50 healthy tissue samples from the GBM tumor margin were collected as a control. RNA extraction and cDNA synthesis were performed for all samples. Subsequently, a specific primer was designed and the expression of SCAMP5 gene was examined using Real-time PCR. ACTB gene was used as the internal control. Statistical analysis was carried out using Graphpad Prism software version 8. Roc curve was used to evaluate the biomarker value of SCAMP5 gene. Decreased expression of SCAMP5 gene was observed in GBM samples and its expression was compared with the pathological characteristics of patients. Expression of SCAMP5 gene with patients’ age with P-value ˂0.05 was statistically significant. However, the expression of this gene with patients’ gender with P-value ˃ 0.05 was not statistically significant. SCAMP5 was expressed in tumor tissue of patients with GBM much lower than healthy tissue around the tumor. The results revealed that SCAMP5 expression decreased in patients older than 50 years. ROC curve showed that the SCAMP5 gene could have a biomarker value. According to this study, it may be possible to use the expression of this gene as a marker as an early diagnostic method.         Glioblastoma multiforme (also called glioblastoma, GBM, or grade 4 astrocytoma) is uncommon cancer which accounts for more than half of the primary malignant brain tumors diagnosed. The study of the expression of genes involved in the glioblastoma can be effective in early detection, so we discussed the clinical significance and biological role of the SMC4 gene transcript in glioblastoma cancer, which is unknown. Before treatment, 50 samples of healthy tissue margin from a glioblastoma tumor were taken as a control group, and 50 samples of glioblastoma tissue were acquired as a patient group. We did RNA extraction and cDNA synthesis. qRT-PCR was used to determine SMC4 expression levels. As an internal control, ACTB was employed. Statistical analysis was performed by GraphPad Prism v.8.0.1. ROC curve was plotted.Overexpression of SMC4 (P<0.0001) in glioblastoma tissue samples was significant compared to healthy tumor margin tissue. While in age (P<0.7507) and sex (P<0.2270) no significant expression was observed. Increased expression of SMC4 gene can play an important role in the glioblastoma cancer. Our work provides a new perspective on SMC4 gene transcription with respect to ROC curve analysis as a biomarker, but needs further study.  Manuscript profile

        Glioblastoma is an aggressive malignant tumor of the brain and spinal cord. Several lines of evidence indicate an important oncogenic role of highly non-coding RNA (lncRNA) molecules in a variety of cancers including glioma, however, the tumorigenic function of lens in glioma remains largely unclear. Increasing evidence has shown that lncRNAs, including LncRNA SNHG15, play an important role in the pathophysiology of human diseases, especially in the pathogenesis and progression of cancers. In this study, 25 paraffin tissue blocks of patients with glioblastoma multiforme and tumor margin tissue were collected and RNA extracted. Synthesis of cDNA and analysis of SNHG15 expression was done by Real-Time PCR technique. ROC curve was drawn to check the biomarker value and statistical analysis was done by GraphPad Prism v.8.0.1. An increase in the expression of SNHG15 was recorded in the tissue samples of patients compared to the tissue of the tumor margin (p < 0.0001). There was a significant relationship in the expression of this gene in patients aged more than 50 years and less than 50 years (p = 0.6573), Frontal, Temporal, Parietal, Occipital, and Other locations (P value = 0.9802), survival in more than 12 months and less than 12 months (p = 0.5007) was not observed, only in male and female sex (p = 0.0001) was registered. The expression of LncRNA SNHG15 was recorded in the tumor tissue of patients with glioblastoma multiforme more than in the peripheral tumor tissue. By examining the ROC curve, it is possible that LncRNA SNHG15 can be proposed as a biomarker, but it needs more studies. Manuscript profile
