        Chicory is used in traditional medicine as a kidney protector and stone breakers. In this study, the effect of chicory stem aqueous extract on the prevention of calcium oxalate stone formation in the kidneys of rats was investigated. 24 male Wistar rats were assigned to four groups as follows: healthy control, received ethylene glycol and prevention groups (administered 50 mg/kg and 200 mg/kg intraperitoneal [i.p.] dose of chicory stem aqueous extract along with 1% ethylene glycol for 30 days. On days 0, 15, and 30, urine volume and the urinary levels of citrate, calcium, uric acid, creatinine, and pH and on day 31, the serum levels of calcium, uric acid, and creatinine were evaluated. RNA was extracted from kidney tissues and cDNA was synthesized. The expression levels of IL-1β, TNF-α -coding genes’ were evaluated by real-time PCR technique. Data analysis was performed using one-way analysis of variance (ANOVA) and Tukey's post hoc test in SPSS. In the prevention groups calcium urinary level (day 15 and dose 50 mg/kg), citrate (day 15) and serum calcium, uric acid, creatinine, as well as TNF-α expression had reduced significantly. Urine volume (day 30) and IL-1β-coding gene’ expression (200 mg/kg) had increased significantly. Cichorium intybus L. stem is probably causing a decrease in serum parameters and urine calcium at the dose of 50 mg/kg due to its diuretic properties. But at the dose of 200 mg/kg, it has no effects on preventing nephrolithiasis and increases renal inflammation caused by increased IL-1β and some urinary parameters. Manuscript profile

        Natural antioxidant compounds can be a suitable substitute in pharmaceutical industries. Callus of (Cichorium intybus L) is a good source for antioxidant production. The research aim is to evaluate the amounts of phenolic compounds, flavonoid and antioxidant activity of plant in vitro and natural conditions. For this purpose, after preparing healthy seedlings from chicory seeds in (1.2 MS) culture medium, stem, leaf and root explants were prepared and cultured for callus formation in liquid MS with different hormone treatments. Methanolic extracts were prepared from each Callus.The phenolic content and flavonoid were determined by Folin – Ciocalteu and aluminum chloride methods and antioxidant activity by measuring the reduction of radical capacity by DPPH. The HPLC method was used to determine the type of phenolic compounds in the extracts.The results showed the highest amount of total phenol is 2.613 mg/g of the Callus weight related to the leaf explant with Kin (0.5mg/L), the highest amount of flavonoid is 0.828 mg/g related to the stem explant with 2,4-D (0.5mg /L) and the highest amount of total antioxidant was 89.35% related to the stem explant with BAP (1mg/L).The highest amount of chlorogenic acid was 0.513 mg/g related to the leaf explant with BAP (0.75 mg/L) and the highest amount of caffeic acid was 0.009 mg/g related to the stem explant with Kin (1 mg/L).The Callus obtained from stem and leaf explants, having a good level of phenolic and flavonoid compounds, can be used as a source of natural antioxidants instead of synthetic antioxidants.         Chicory (Cichorium intybus L) is an important medicinal plant with valuable secondary metabolites. Chicory callus is a good source for the production and extraction of metabolites of this plant. In this study, chicory callus changes were investigated about some morphological and biochemical characteristics to obtain the best hormonal treatment. Healthy seedlings were prepared from sterile chicory seeds in ½ MS culture medium. Stem, leaf, and root explants were prepared from seedlings and used for culture in liquid MS and 2,4-D, BAP, and kinetin hormones at concentrations of 0.5, 0.75, and 1 mg / l were cultured. The results showed that callus was yellow and green to dark brown and callus texture also showed from weak and soft to firm. The highest percentage of callus (76.22%) related to leaf explant and treatment 2,4-D (0.5mg / L), the highest amount of fresh weight of callus (7.30 g), and dry weight of callus (0.077 g) In leaf explant and Kin treatment (0.75 mg / L), the highest amount of chlorophyll a, b and total was related to leaf and BAP hormone treatment (0.75 mg / L). The highest amount of carotenoids was related to root explant and Kin (1 mg / L). In general, the interactions of hormones and explants affect different callus-forming characteristics and these effects depend on the type of explants and the concentration of growth regulators, and the greatest effect was observed in leaf explant culture.
