1 - بررسی اثر کلسیم بر روی رشد، شاخصهای اکسیداتیو، آسکوربات، گلوتاتیون و فعالیت آنزیمهای آنتیاکسیدان در گیاه تریتیکاله (Triticosecale Wittmack ×) تحت تنش شوریIranian Journal of Biological Sciences , Issue 2 , Year , Autumn 2022مقدمه: شوری یک مشکل در حال توسعه در خاکهای کشاورزی است. کلسیم نقش مهمی در مقاومت گیاهان به تنش شوری دارد. هدف: به منظور بررسی تأثیر متقابل سدیم-کلسیم بر روی رشد، شاخصهای اکسیداتیو و سیستم دفاع آنتیاکسیدانی در گیاه تریتیکاله، آزمایشی بصورت یک طرح کاملاً تصادفی با 3 تک Moreمقدمه: شوری یک مشکل در حال توسعه در خاکهای کشاورزی است. کلسیم نقش مهمی در مقاومت گیاهان به تنش شوری دارد. هدف: به منظور بررسی تأثیر متقابل سدیم-کلسیم بر روی رشد، شاخصهای اکسیداتیو و سیستم دفاع آنتیاکسیدانی در گیاه تریتیکاله، آزمایشی بصورت یک طرح کاملاً تصادفی با 3 تکرار در شرایط گلخانهای صورت گرفت. مواد و روشها: یک هفته بعد از کاشت بذور در خاک، گیاهچهها با غلظتهای کلرید سدیم (0، 50، 100 و 150 میلیمول بر لیتر) و توأم با غلظتهای کلرید کلسیم (0، 6 و 10 میلیمول بر لیتر) تیمار شدند. پس از 5 هفته اعمال تنش، برخی از پارامترهای مورفو- فیزیولوژیکی و بیوشیمیایی شامل طول بخش هوایی و ریشه، عدد کلروفیلمتر (SPAD)، شاخصهای اکسیداتیو (مالوندیآلدئید، سایر آلدئیدها، پراکسید هیدروژن و فعالیت آنزیم لیپواکسیژناز)، آسکوربات احیا، دهیدروآسکوربات، گلوتاتیون احیا و فعالیت آنزیمهای آنتیاکسیدان (گایاکول پراکسیداز، کاتالاز، سوپراکسید دیسموتاز، آسکوربات پراکسیداز و پلیفنل اکسیداز) برگ مورد سنجش قرار گرفت. نتایج: نتایج نشان داد که تنش شوری بطور معنیداری باعث کاهش طول بخش هوایی و ریشه، SPAD، آسکوربات احیا و گلوتاتیون احیا در برگ شد ولی باعث افزایش میزان شاخصهای اکسیداتیو، دهیدروآسکوربات و فعالیت آنزیمهای آنتی اکسیدان برگ شد. درحالیکه افزودن کلسیم به محیط شور باعث افزایش طول بخش هوایی و ریشه، SPAD، آسکوربات احیا و گلوتاتیون احیا در برگ شد ولی باعث کاهش میزان شاخصهای اکسیداتیو، دهیدروآسکوربات و فعالیت آنزیمهای آنتی اکسیدان در برگ شد. نتیجهگیری: افزودن کلسیم به محیط شور باعث کاهش اثرات مضر تنش شوری شد و بیشترین اثرات بهبوددهنده کلسیم در غلظت 6 میلیمول بر لیتر مشاهده شد. Calcium has a crucial role in plant resistance to salinity stress. In order to investigate the impact of calcium supplementation on photosynthetic pigments, compatible osmolytes contents and membrane stability index (MSI) in triticale (x Triticosecale Wittmack) exposed to salinity stress, an experiment as a completely randomized design with 3 replications in greenhouse condition (25 ± 2°C, 35% relative humidity, 16-hour photoperiod) was conducted. The seeds were germinated in soil. One week old triticale seedlings (with two leaves) were imposed by 0, 50, 100 and 150 mmol L-1 NaCl and 0, 6 and 10 mmol L-1 CaCl2 for 5 weeks and assayed for some morpho-physiological parameters including fresh weight (FW) and dry weight (DW) of shoot, photosynthetic pigments (chlorophyll (Chl) a and Chl b, total Chl and carotenoids (Car)) contents, proline and glycine betaine (GB) contents, soluble sugars and starch contents and MSI in leaves. Results showed that with incrementing salinity meaningfully decremented FW and DW of shoot, photosynthetic pigments, starch content and MSI while proline, GB and soluble sugars contents incremented in leaves. Calcium treatment meaningfully incremented FW and DW of shoot, photosynthetic pigments, starch content and MSI but caused a meaningful decline in proline, GB and soluble sugars contents in leaves. It can be concluded that calcium had exerted an ameliorative impact on triticale under salinity stress. Maximum ameliorative impact of calcium was observed in plants exposed to 6 mmol L-1 CaCl2.
