اثر پیوند سلول بنیادی مغز استخوان به همراه انجام یک دوره تمرین هوازی بر برخی شاخص¬های التهابی و آسیب قلبی در موش¬های صحرایی نر به دنبال القاء سکته قلبی

زمینه و هدف: سکته قلبی یکی از شایع¬ترین بیماری های ایسکمیک قلبی است که امروزه روش¬های درمانی جدیدی برای کنترل و درمان آن مورد توجه قرار گرفته است. هدف پژوهش حاضر، بررسی اثر پیوند سلول بنیادی مغز استخوان به همراه انجام یک دوره تمرین هوازی بر شاخص¬های التهابی و آسیب قلبی در موش¬های صحرایی نر به دنبال القاء سکته قلبی بود. مواد و روش¬ها: 60 سر موش نر در یک مطالعه تجربي به 6 گروه 10 تایی کنترل سالم، بیمار، شم، تمرین، سلول و تمرین + سلول تقسیم شدند. برای القاء سکته قلبی، شریان پایین رونده قدامی با استفاده از نخ سیلک 6/0 به طور کامل مسدود شد. سلول بنیادی مغز استخوان با استفاده از لام نئوبار، حدود 106 سلول برای هر موش از طریق سیاهرگ دمی تزریق شد. موش¬های گروه تمرینی به مدت 10 هفته تمرین هوازی را با شدت 17 متر بر دقیقه انجام دادند. شاخص¬های پژوهش به وسیله کیت¬ آزمایشگاهی اندازه-گیری شد نتایج: مقادیر TNF-α و IL-10 در گروه¬های تجربی نسبت به گروه بیمار به ترتیب کاهش و افزایش یافت اما تغییرات TNF-α تنها در گروه تمرین + سلول و تغییرات IL-10 تنها در گروه تمرین و تمرین+سلول نسبت به گروه بیمار معنی¬دار بود. شاخص¬های آسیب CK و LDH در گروه¬های تجربی تمرین، سلول و تمرین + سلول نسبت به گروه بیمار کاهش معنی¬داری یافت. نتیجه¬گیری: پیوند سلول بنیادی مغز استخوان به همراه انجام یک دوره تمرین هوازی سبب کاهش التهاب و کاهش شاخص¬های آسیب قلبی در موش¬های مبتلا به سکته قلبی گردید.
Introduction: Cardiac infarction is one of the most common ischemic heart diseases, and new treatment methods are being considered for its control and treatment. The aim of the present study was to investigate the effect of bone marrow stem cell transplantation with an aerobic training course on inflammatory and cardiac damage indices in male rats with myocardial infarction. Methods: In an experimental study, 60 male rats were divided into 6 groups of 10 rats: healthy control, patient control, sham, training, cell, and training + cell. To induce myocardial infarction, the anterior descending artery was completely blocked using 0.6 silk thread. Bone marrow stem cell extraction, using neobar slide, about 106 cells were injected for each mouse through the tail vein. Rats in the training group performed aerobic exercise with an intensity of 17 m/min for 10 weeks. The research indicators were measured by laboratory kit. Results: The serum levels of TNF-α and IL-10 in the experimental groups decreased and increased, respectively, compared to the patient group, but the changes of TNF-α only in the training + cell group and the changes of IL-10 only in the training and training + cell groups compared to the patient group was significant. CK and LDH damage indices were significantly decreased in the experimental groups of exercise, cell and exercise + cell compared to the patient group. Conclusion:In the present study, bone marrow stem cell transplantation with a course of aerobic training reduced inflammation and reduced heart damage indicators in rats suffering from heart attack.
