بررسی هیستومورفولوژی کلیه و برخی شاخص های خون شناسی تیلاپیای نیل (Oreochromis niloticus) طی جایگزینی پودر خون بجاي پودر ماهی در جیره غذایی

مطالعه حاضر با هدف تأثیر جایگزینی پودر خون بهجاي پودر ماهی بر هیستومورفولوژی بافت کلیه و برخی شاخصهای خونی تیلاپیای نیل صورت گرفت. ماهیان تیلاپیا پس از آمادهسازی به مدت ۸ هفته با جیرههای طراحی شده شامل کنترل، 25، 50، 75 و100 درصد پودر خون تغذیه شدند. پس از این مدت ماهیان بیهوش شده و خونگیری از ساقه دمی با سرنگ هپارینه انجام گرفت. ضمن باز کردن محوطه شکمی نمونههای به ابعاد پنج میلیمتر از کلیه برداشت شد. پس از قرار دادن در محلول ثبوتی فرمالین بافر ۱۰ درصد و پس از طی مراحل آبگیری با الکل، شفافسازی با گزیلول، بلوکگیری با پارافین، برشگیری به ضخامت 6-4 میکرون با میکروتوم انجام گرفت. در ادامه ضمن رنگآمیزی هماتوکسیلین-ائوزین و درنهایت مطالعه با میکروسکوپ نوری بر روی آنها انجام گرفت. نتایج نشان داد که در گروه دریافت کننده پودر خون تغییرات متعددی مانند تجمع هیالن، کنده شدن اپیتلیوم، اتساع کپسول بومن، پرخونی، افزایش مراکز ملانوماکروفاژ و نکروز گلومرول و توبولها مشاهده گردید که هرچه بر مقدار درصد پودر خون جیره افزوده شده بر شدت ضایعات وارده نیز افزوده گردید. با افزایش میزان جایگزینی پودرخون بجاي پودرماهی در جیره تعداد گلبولهای قرمز، درصد هماتوکریت، میزان هموگلوبین خون روند کاهشی و تعداد لنفوسیتها، مونوسیت و نوتروفیلها با افزایش سطح پودر خون روند افزایشی نشان دادند و بین تیمارها بویژه در سطوح بالا اختلاف معنیدار مشاهده شد (۰۵/۰p <). بنابر نتایج این تحقیق پودر خون در جیره غذایی تا حدود 75 درصد برای گونه مورد نظر قابل تحمل و جایگزینی در جیره میباشد.
The present study aimed to investigate the effect of replacing blood powder with fish powder on the histomorphology of kidney tissue and some blood indices of Nile tilapia. After preparation, the fish were fed with designed diets for 8 weeks, including control, 25, 50, 75, and 100% blood powder. After this period, the fish were anesthetized and blood was collected from the caudal peduncle with a heparin syringe. While opening the abdominal cavity, five-millimeter samples were taken from the kidney. After placing in a 10% buffered formalin fixative solution and undergoing dehydration with alcohol, clarification with xylene, and paraffin blocking, sections were made to a thickness of 4-6 microns with a microtome. They were then stained with hematoxylin-eosin and finally studied with a light microscope. Results showed that in the group receiving blood powder, numerous complications such as hyaline accumulation, epithelial rupture, Bowman's capsule dilation, hyperemia, increased melanomacrophage centers, and glomerular necrosis and tubules were observed. As the percentage of blood powder in the diet increased, so did the severity of the lesions. With increasing the replacement of blood powder instead of fish powder in the diet, the number of red blood cells, the percentage of hematocrit, and the amount of hemoglobin show a decreasing trend. The number of lymphocytes, monocytes, and neutrophils increased with increasing levels of blood powder. There was a significant difference between treatments, especially at high levels (p < 0.05). According to the results of this study, blood powder can be tolerated and replaced up to 75% of the diet for this species.
