    Calpastatin gene has been known as a candidate gene for meat quality in cattle, sheep, and chicken. The purpose of this study was to identify CAST gene polymorphisms and its association with meat traits in Boerka goat. The data of pH, cooking loss (CL), warner-bratzler shear force (WBSF), water holding capacity (WHC), cholesterol, water, ash, fat, and protein contains were recorded. Sequencing of 21 samples revealed five polymorphisms of CAST gene in intron 12 within Boerka goat, namely g.146C > A, g.224A > G, g.281G > A, g.737C > T, and g.431G > A. Only single nucleotide polymorphism (SNP) g.281G > A was used for genotyping. The genotype and allele frequency based on g.281G > A showed 14.29% (GG genotype) and 85.71% (GA genotype) followed with 57% G allele and 43% A allele. The chi-square test showed deviation from HWE (p <0.05) in Boerka goat. The SNP g.281G > A revealed having significantly effect to CL. The GA animals had lower CL percentage compared to the GG animals. In conclusion, the SNP selected may be used for identify meat having low CL in Boerka goat. تفاصيل المقالة

    Melanocortin-4 receptor (MC4R) gene has been known as a candidate gene for growth traits in livestock. This research was aimed to identify the polymorphism of MC4R gene and its association with growth traits in Sumba Ongole (SO) cattle. The growth traits data consist of body weight and body size. Eighty four blood samples were collected and used for DNA isolation. The results showed one SNP (g.1133C&gt;G) resulting in two alleles (C and G) and three genotypes (CC, CG, and GG) in SO cattle. The frequency of G allele (0.59) was higher than C allele (0.41). The CG genotype had the highest value of genotype frequency (0.48). The allele and genotype distributions followed the Hardy-Weinberg equilibrium. The single nucleotide polymorphism (SNP) g.1133C &gt; G revealed having no significant association with growth traits in SO cattle. In conclusion, the SNP g.1133C &gt; G may not useful as selection tool in Sumba Ongole cattle. It can be suggested that the future study is needed to find out the effect of the MC4R gene in Sumba Ongole cattle in different locus. تفاصيل المقالة

    The melanocortin-1 receptor (MC1R) gene has been investigated by many studies regarding the pigmentation variation in various species. In order to determine its allelic and genotypic distribution, we sequenced the goat MC1R gene from 78 individuals in ten populations (Gembrong, Senduro, Ettawa Grade, Boerawa, Boerka, Kosta, Samosir, Muara, Boer and Kacang). Direct sequencing method was performed to detect the single nucleotide polymorphisms (SNPs). Three SNPs (g.676A&gt;G, g.748G&gt;T and g.801C&gt;G) were identified in the gene target. The SNP g.676 A &gt; G was used to genotype the investigated animals by using polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) method with EarI restriction enzyme. In all studied populations, the A allele had the highest frequency (83%) and the G allele had the lowest frequency (17%). The frequency of homozygous AA genotype was the highest (90%) in Kacang goats, while that of heterozygous AG genotype was fixed in Senduro and Boer goats. Analysis of observed heterozygosity, expected heterozygosity, Nei&rsquo;s (1973) expected heterozygosity, and Shannon index were found to be 0.3425, 0.2858, 0.2838 and 0.4578, respectively.The distribution of MC1R gene in overall breeds confirmed with Hardy-Weinberg equilibrium (P&gt;0.05). تفاصيل المقالة
