1 - بهینهسازی شرایط PCR برای دو ژن CHD-W و CHD-Z جهت تفکیک جنسیت در قناری
فاطمه اعتمادی، زینب فتحخانی، احمد ابراهیمی، خسرو حسینیپژوه .در گونههای بسیاری از پرندگان من جمله قناریها، جنس نر و ماده قبل از بلوغ فاقد هر گونه تفاوت در شکل ظاهری میباشند. از آنجاییکه صدای دلنشین قناری توسط جنس نر ایجاد میشود، لذا تفکیک جنسیّت قناریها قبل از بلوغ از لحاظ اقتصادی بسیار حائز اهمّیّت میباشد. ژنهای موجود بر چکیده کاملدر گونههای بسیاری از پرندگان من جمله قناریها، جنس نر و ماده قبل از بلوغ فاقد هر گونه تفاوت در شکل ظاهری میباشند. از آنجاییکه صدای دلنشین قناری توسط جنس نر ایجاد میشود، لذا تفکیک جنسیّت قناریها قبل از بلوغ از لحاظ اقتصادی بسیار حائز اهمّیّت میباشد. ژنهای موجود بر کروموزومهای جنسی به خاطر منحصر به جنس بودنشان میتوانند برای تفیین جنسیت به روش مولکولی مورد استفاده قرار گیرند. با استفاده از روش PCR و بر اساس دو ژن CHD-W و CHD-Z تفکیک جنسیّت در قناریها صورت گرفته است. هدف از این مطالعه بهینه سازی PCR بر اساس دو ژن CHD-W و CHD-Z در تفکیک جنسیّت در قناری بود. در این مطالعه از 24 قناری دو پر حاوی ریشه تهیه گردید و استخراج DNA از بالب پرها صورت پذیرفت و از پرایمرهای اختصاصی طراحی شده برای تکثیر توالی ژن W-CHD استفاده شد و با بهینه سازی شرایط PCR برنامه بهینه به صورت زیر به دست آمد: واسرشته سازی اوّلیه 94 درجه سانتیگراد به مدّت 5 دقیقه، 35 سیکل با شرایط : 94 درجه سانتیگراد به مدّت 60 ثانیه ،58 درجه سانتیگراد به مدّت 35 ثانیه و دمای 72 درجه سانتیگراد به مدّت 50 ثانیه و در نهایـت دمـای72 درجه سانتیگراد بـه مـدت 5 دقیقه. این مطالعه نشان داده شد که روش بهینه شده PCR میتواند با دقت صد درصد جنسیت قناری را تعیین کند و لذا مناسبترین روش برای تعیین جنسیّت قناری است. پرونده مقاله -

2 - Some Parts of the Feather Can be a Non-Invasive Genetic Sample for Sexing in Avian?

The purpose of this study was to evaluate DNA extracted from 7 different parts of the feather (calamus tip, rachis (I-III) and barbs (I-III)) for sex identification using polymerase chain reaction (PCR) amplification. The study results showed that the calamus tip had the highest DNA concentration. DNA extracted from rachis and barbs can also be amplified and use for sex determination. Extracted DNA from the calamus tip, rachis (I), and barbs (I) showed accurate results for amplified PCR products when compared to the sex of known samples. The findings revealed that, in addition to the calamus tip, the rachis and barbs on the lower part of the feather surrounding on the calamus can be used to determine sex. Calamus tip collection by plucking is an invasive technique that could result in contusion and infection. The non-invasive sample collection method of cutting a portion of the feather is one option for supporting animal welfare guidelines. Furthermore, the non-invasive method presented in the study can be used to collect samples in other branches of molecular biology. پرونده مقاله
