تاثیر میکروگراویتی بر رشد، فراساختار، میزان فنل کل، فلاونوئید، و مهار رادیکال DPPH در دانهرستهای گیاه بذرالبنج (.Hyoscyamus niger L)

چکیدهمیکروگراویتی به عنوان یکی از تنشهای محیط فضا میتواند روی رشد اندامهای گیاه، ساختار اندامکهای سلولی و متابولیتهای سلول تاثیر بگذارد. در پژوهش حاضر تاثیر میکروگراویتی به مدت 3، 7 و 10 روز بر میزان رشد هیپوکوتیل و ریشهچه، وزنتر و خشک، فراساختار سلول و اندامکها با استفاده از میکروسکوپ الکترونی و همچنین ترکیبات آنتیاکسیدانی نظیر فنل کل، فلاونوئید و مهار رادیکال DPPH در دانهرست گیاه بذرالبنج (Hyoscyamus niger L.) مورد بررسی قرار گرفت. نتایج نشان داد تنش میکروگراویتی موجب افزایش طول ریشهچه و هیپوکوتیل گردید در حالیکه روی وزن تر و خشک نسبت به شاهد اختلاف معنیداری نشان نداد. دانههای نشاسته در تیمار میکروگراویتی در تمام سلول پراکنده بودند. در حالیکه در نمونه کنترل در جهت جاذبه روی دیواره سلول رسوب یافتند. تعداد میتوکندری و شکل آن در تیمارها متغیر بود. شبکه آندوپلاسمی در تیمار 10 روز حجیمتر و بلندتر از شاهد مشاهده شد. واکوئل تحتتاثیر میکروگراویتی حجیمتر شد. میزان فنل کل، فلاونوئید و فعالیت مهار رادیکال DPPH تحت تیمار میکروگراویتی نسبت به شاهد اختلاف معنیداری نشان داد ( P < 0.05). بنظر میرسد تغییرات سلولی و متابولیسم ثانوی القا شده تحت میکروگراویتی در ارتباط با ایجاد تنش اکسیداتیو باشد.

AbstractMicrogravity is one of the environmental stresses in space that affects on the structure of cellular organelles and metabolites in plant. In the present research, the effect of microgravity in times 3, 7 and 10 days on root and hypocotyl growth rate, fresh and dry weight, cell and organelles ultrastructure with Transmission Electron microscope (TEM) and some antioxidative capacity ( phenol, flavonoid and inhibition of DPPH radicals) were investigated in henbane (Hyoscyamus niger L.) seedling. The results showed that microgravity led to increase root and hypocotyle length as compared to control. Amydons were spread throughout the cell in microgravity treatment, while in control samples, they were deposited on the cell wall in the direction of gravity. Mitochondria were variable in shape and number. The endoplasmic reticulum in 10-day treatment was more voluminous and longer than the control. Also, the vacuole was observed more voluminous as compared to control. Total phenol, flavonoid, and inhibition of DPPH radicals increased significantly(P < 0.05). It seems, cellular changes and secondary metabolism induced under microgravity are related to oxidative stress.
