بررسی اثر عصاره هیدرو الکلی بذر گیاه زنیان (Trachyspermum Copticum) بر رفتار اضطراب در موش های صحرایی نر بالغ

چکیده مقاله :
اضطراب یک اختلال شایع است که با یک احساس ناخوشایند با منشاء ناشناخته همراه است. با توجه به اثرات ضداضطرابی زنیان در طب سنتی، در این پژوهش تاثیر بذر زنیان بر اضطراب موش های نر نژاد ویستار مورد بررسی قرار گرفته است. پس از شناسایی بذر گیاه زنیان توسط هرباریوم دانشگاه، عصاره هیدرو الکلی توسط سوکسله استخراج و با استفاده از آون هوشمند، پودر عصاره تهیه گردید. در این مطالعه تجربی موش های صحرایی نر نژاد ویستار (20 ± 230 گرم) در گروه های کنترل، دریافت کننده ی حلال عصاره یا دی متیل سولفاکساید (3/0 سی سی) و دریافت کننده دوزهای 15، 25، 50 میلی گرم بر کیلوگرم عصاره هیدرو ا لکلی زنیان قرار گرفتند. تمامی تزریقات به صورت درون صفاقی انجام گرفت. نیم ساعت بعد از تزریق، با استفاده از ماز صلیبی شکل مرتفع، آزمون اضطراب گرفته شد و به مدت 5 دقیقه شاخص های استاندارد ارزیابی اضطراب (مدت زمان ماندن و تعداد دفعات ورود به بازوی باز) بررسی و ثبت شد. داده ها توسط آنالیز واریانس یکطرفه و تست توکی مورد تجزیه و تحلیل قرار گرفتند. نتایج نشان داد گروه دریافت کننده حلال عصاره نسبت به گروه کنترل از نظر درصد مدت حضور و تعداد دفعات ورودبه بازوی باز تفاوت معنی داری را نشان نمی دهد. گروه دریافت کننده دوز 15 میلی گرم بر کیلوگرم عصاره نسبت به گروه دریافت کننده حلال عصاره از نظر مدت حضور و تعداد دفعات ورود به بازوی باز افزایش معنی داری را نشان داد (001/0 > p). عصاره هیدرو الکلی زنیان دردوز پایین اثرات ضداضطرابی دارد.

Anxiety is a common disorder of unknown origin is associated with an unpleasant feeling. According to the anxiolytic effects TC traditional medicine, Trachyspermum copticum effect on anxiety male Wistar rats was investigated. T. copticum seed after detection by the university herbarium, hydro-alcoholic extract was extracted by Soxhlet and the extract powder was prepared using smart legitimate. Male Wistar rats (230 ± 20 gr) were placed. In groups intact, recipient of the DMSO (solvent extraction) (0.3 ml), received extract in different doses of 50, 25, 15 mg/kg. All injections were performed intraperitoneally. Half an hour after injection, anxiety was measured using the elevated plus maze. Indicators of anxiety for 5 min, including time spent and number of entries into the open arm was recorded. Data were analyzed by ANOVA statistical tests and Tukey test. The group receiving the extract solvent (DMSO) compared to intact in time spent and number of entries into the open arm did not show a significant difference. The group receiving a dose of 15 mg/kg compared to the group receiving DMSO in time spent and number of entries into the open arm showed a significant increase. T. copticum hydroalcoholic extract has anxiolytic effects in low doses.
