Optimization of in vitro proliferation of medicinal herb (Nasturtium officinale)

Abstract :
Nasturtium is of particular importance in terms of medicinal and nutritional aspects. A factorial experiment was conducted in a completely randomized design with five replications and three observations in three separate experiments (seed germination, proliferation and rooting). The germination treatments included temperature (5, 10, 15 and 25 °C) and time (7, 14, 21 and 28 days) and growth regulator treatments including BA and NAA, NAA and TDZ, and NAA and Kin, that BA, TDZ and Kin at concentrations of 0, 0.5, 1 and 2 mg L-1 and NAA at concentrations of 0, 0.01, 0.02 and 0.04 mg L-1 in proliferation and IBA at concentrations of 0, 0.25 and 0.5 mg L-1 in rooting. The results showed that the number of germinated seeds and the rate of germination increased during time, at high temperatures, especially at 15°C. In proliferation, the highest seedling length was obtained by the combined application of NAA (0.04 mg L-1) and kinetin (2 mg L-1) had a significant difference with other treatments. The highest number of leaves was obtained in TDZ treatment (2 mg L-1) and combined with a concentration of 0.44 mg L-1 NAA. The maximum length of the shoots in the Kinetin (1 mg L-1) and in combination with a concentration of 0 mg L-1 NAA. In rooting, with increasing concentration of IBA, the number and length of the formed roots were increased and the concentrations of 0.5 mg L-1 resulted in the best seedling response to rooting. In experiments on proliferation, the highest seedling length was obtained by the combined application of NAA at a concentration of 0.04 mg L-1 and kinetin at a concentration of 2 mg L-1, which produced a significant difference with other treatments.
