In-vitro effect of iron sulfate, zinc sulfate and copper sulfate treatments on the morphophytochemical characteristics of Chamaecostus cuspidatus Nak

Abstract :
In order to investigate the effects of changing the concentration of iron sulfate, zinc sulfate, and copper sulfate micronutrients in MS medium on the morphological and phytochemical factors of the insulin plant, an experiment was conducted based on a completely randomized design in the tissue culture laboratory of the Faculty of Agriculture of University of Zanjan. The nodal explants were grown for 12 weeks in MS culture medium with a hormone concentration of 1 mg/L-1 benzyl adenine and 0.6 mg/L-1 naphthalene acetic acid. After 12 weeks, the explants were treated for 4 weeks with different levels of iron sulfate (0, 27.8, 41.7, 55.6 mg/L-1), zinc sulfate (0, 8.6, 17.2, 25.8 mg/L-1) and sulfate Copper (0, 0.025, 0.05, 0.1 mg/L-1) were placed. After 4 weeks, growth and phytochemical indices were evaluated. The results of the experiment showed that the effect of the treatments on the studied traits was significant at 1% probability level. According to the average data comparison results, copper sulfate with a concentration of 0.1 mg/L-1 caused the highest number of leaves (7.33), shoot height (9.20 cm), root length (14.67 cm) and fresh weight (26.1 g) and shoot dry weight (180 mg), fresh weight (0.58 g) and root dry weight (146.67 mg). A significant increase in chlorophyll a and total chlorophyll was obtained in zinc sulfate with a concentration of 25.8 mg/L-1 and copper sulfate with a concentration of 0.1 mg/L-1. The highest total carotenoid was obtained at the concentration of 17.2 zinc sulfate and 0.1 mg of copper sulfate. In conclusion, the concentration of 0.1 mg/L of copper sulfate had the greatest effect on morphological traits and photosynthetic pigments, and the levels of 17.2 mg/L-1 of zinc sulfate and 0.05 mg/L-1 of copper sulfate had a significant increase in total phenolic content and antioxidant activity.
