بررسی اثر ضدقارچی اسانس چهار گیاه دارویی، آنالیز و شناسایی ملکولی باکتری آنتاگونیست علیه قارچ عامل پوسیدگی بعد از برداشت پرتقال Penicillium digitatum

ملخص المقالة :
کپک سبز مرکبات Penicillium digitatumازمهمترین بیماریهای بعد از برداشت میباشد. به کارگیری مداوم سموم شیمیایی تاثیر ماندگار روی سلامتی انسان و آلودگی محیط زیست دارد و سبب بروز مقاومت در سویههای بیماریزا می گردد. در این تحقیق فعالیت ضدقارچی غلظتهای مختلف (100، 200، 300 و 400 پیپیام) اسانس چهار گیاه دارویی آویشن شیرازی (Thymus vulgaris)، آلوورا (Aloe vera)، سیر (Allium sativum) و زیره سبز (Cuminum cyminum) جمعآوری شده از منطقه قرق استان گلستان مورد بررسی قرار گرفت. اسانسها با استفاده از روش تقطیر؛ بخار آب و با استفاده از دستگاه کلونجر تهیه و سپس مهمترین مواد موثره اسانسی که بیشترین عملکرد آنتاگونیستی را داشتند توسط دستگاه GC/MS آنالیز گردید. همچنین فعالیت آنتاگونیستی باکتریهای جداسازی شده از اپیفیت پرتقال شامل sp. Bacillus، Pseudomonassp.، Streptomycess sp. علیه قارچ پنی سیلیوم در قالب طرح فاکتوریل مورد بررسی قرار گرفت. بیشترین درصد ممانعت کنندگی (95/99 درصد)، توسط اسانس آویشن علیه پنی سیلیوم در غلظت 400 پیپیام مشاهده گردید. آنالیز رگرسیونی تاثیر اسانسها نشان داد با افزایش غلظت اسانس درصد بازدارندگی آن روی قارچ افزایش مییابد. بر اساس نتایج به دست آمده، اسانس آویشن با داشتن مهمترین ترکیبات موثره در اسانسها (کارواکرول - 5/45 درصد)، آلفا- ترپینئول (-9/22 درصد) و اندو- بورنئول (3/14 درصد) بیشترین فعالیت ممانعت کنندگی (3/22 میلیمتر) را علیه پنی سیلیوم از خود نشان داد. در بین 23 جدایه باکتریایی، یک جدایه به نام H7 بیشترین فعالیت ممانعت کنندگی را علیه قارچ داشت. با استفاده از PCR و تعیین توالی DNA ژنوم 16S rRNA، H7 بهعنوان Bacillus cereus شناسایی گردید. بنابراین اسانس آویشن و جدایه (Bacillus cereus (H7 جایگزینهای مناسبی برای کنترل شیمیایی پوسیدگی بعد از برداشت پرتقال می باشند.
