ارزیابی اثر مهارکنندگی عصارههای خیار دریایی (Holothuria parva) بر آنزیم آلفا آمیلاز

ملخص المقالة :
مهارکنندههای آنزیم آلفا آمیلاز از تبدیل پلیساکاریدها به مونوساکاریدها پیشگیری میکند. مهار این آنزیم از جذب قندهای ساده دستگاه گوارش و در نتیجه پیشگیری از افزایش سطح گلوکز خون موثر بوده است. هدف از مطالعه حاضر، بررسی تاثیر عصارهای اتیل استات، متانول و آب- متانول از خیار دریایی Holothuria parva بر مهار آنزیم آلفا آمیلاز است. در این پژوهش از پوسته و امعاء و احشاء گونه Holothuria parva ابتدا عصارهگیری براساس افزایش قطبیت انجام گرفت و در مرحله بعد فعالیت آنزیم آلفا آمیلاز در حضور عصارههای مختلف تحت شرایط سنجش فعالیت آنزیمی به روش رنگسنجی DNSA تعیین شد و IC50 (غلظتی از عصارهها که برای برای مهار 50 درصد فعالیت آنزیم مورد نظر بوده است) بدست آمده و با مقدار آکاربوز به عنوان کنترل مثبت مقایسه شد. در این بررسی نتایج نشان داد که با افزایش غلظت عصارهها درصد مهارکنندگی افزایش یافته و در بین تمام عصارهها بیشترین درصد مهارکنندگی مربوط به آکاربوز در غلظت 8/0 میلیگرم/میلیلیتر برابر 92/96 درصد و کمترین مقدار آن مربوط به عصاره آب–متانول پوسته در غلظت 2/0 میلیگرم/میلیلیتر برابر با 79/61 درصد بوده و درصد بازدارندگی آکاربوز در هر دو اجزای بدنی نسبت به سایر عصارهها بالاتر است (IC50 برابر با 18/88- 74/91 میلیگرم/میلیلیتر). با توجه به اثر مهاری عصارههای استخراج شده از گونه Holothuria parva بر آنزیم آلفا آمیلاز در آینده میتوان در زمینه تولید داروهای ضددیابتی با منشا دریایی با حداقل عوارض جانبی نامطلوب استفاده نمود.
