ملخص المقالة :
سابقه و هدف: روغن حاصل از میکروارگانیسم ها(SCO) نسبت به روغن های گیاهی ارجحیت دارد زیرا حاوی مقادیر بیشتری گامالینولئیک اسید است، پایداری بیشتری در برابر اکسیداسیون و محتوای کمتری از باقیمانده آفت کش ها دارد. این روغن ها به عنوان مکمل های غذایی قابل استفاده هستند. هدف از مطالعه حاضر شناسایی مولکولی مخمر ها و کپک های مولد چربی موجود در خاک و بهینه سازی تولید چربی توسط کاندیدا گلابراتا با طراحی تاگوچی است.مواد و روش ها: از خاک باغات و مناطق نزدیک به مغازه های تعویض روغن و رستوران، نمونه گیری و شناسایی مخمر ها و کپک های تولید کننده ی چربی با روش واکنش زنجیره ای پلیمراز انجام شد. سپس جدایه قارچ با بیشترین میزان تولید لیپید انتخاب و با استفاده از طراحی تاگوچی، بهترین شرایط از نظر میزان منبع کربن و نیتروژن وpH برای تولید حداکثری تعیین گردید. چربی حاصل توسط کروماتوگرافی گازی با آشکارساز طیفسنج جرمی(GC-MS) مورد بررسی قرار گرفت.یافته ها: در بین گونه های شناسایی شده،Candida glabrata strain DHP14-2 بیشترین مقدار چربی را تولید کرد. تولید پالمیتولئیک، پالمیتیک، لینولئیک، لینولنیک و استئاریک اسید در این بررسی ثابت شد. تولید لیپید در کاندیدا گلابراتا معادل 9/6 گرم در لیتر و در مورتیرلا آلپینا معادل 10/8 گرم در لیتر به دست آمد.نتیجه گیری: با توجه به سرعت تکثیر مخمر ها و توانایی تولید اسید های چرب توسط آن ها، کاندیدا گلابراتا گزینه مناسبی برای تولید چربی می باشد.
