چکیده مقاله :
زمینه و هدف: نمدار از جمله گونه‎های مورد تهدید در شمال ایران است که درصد بسیار ناچیزی از جنگل‎های شمال کشور را تشکیل می‎دهد. از آن‎جایی‎که بذور نمدار در جوانه‎زنی با مشکلاتی هم‎چون خواب دوگانه فیزیولوژیکی (جنین) و مکانیکی (پوشش سخت پریکارپ) روبه‎رو هستند، استفاده از روش‎های کاربردی نظیر کشت بافت می‎تواند در کشت، بازسازی و حفاظت از ذخایر ژنتیکی موثر واقع شود. روش بررسی: این پژوهش با هدف تعیین مناسب‎ترین ریزنمونه و بهترین ترکیب هورمونی محیط کشت به منظور بهینه‎سازی ریزازدیادی گونه نمدار در شرایط درون شیشه‎ای انجام گرفت. ریزنمونه‎ها به صورت تصادفی از بهترین پایه‎های نمدار در جنگل توسکاستان گرگان جمع‎آوری و کشت گردیدند. یافته‎ها: داده‎های به ‎دست آمده با طرح کاملا تصادفی به وسیله نرم افزار MSTATC مورد تجزیه و تحلیل قرار گرفت. با توجه به نتایج حاصل از این بررسی، از بین یازده ریزنمونه مورد آزمایش، ساقه حاوی جوانه انتهایی فلس‎دار به عنوان مناسب‎ترین ریزنمونه و IBA با غلظت 1/0 میلی‎گرم بر لیتر به عنوان مناسب‎ترین ترکیب هورمونی محیط کشت در ساقه‎زایی در نظر گرفته شد. بحث و نتیجه‎گیری: امید می‎رود نتایج حاصله بتواند در تحقیقات کشت بافت و تکثیر و هم‎چنین در مطالعات پایه مولکولی در این گونه مفید واقع شود
Background and: Linden is one of threatened species in north of Iran that form very small percentage of the northern forests. Since the linden seeds are facing with germination problems such as with dual physiological (the embryo) and mechanical (hard cover pericarp) dormancy that application such as tissue culture can be effective in planting, restoration and conservation of genetic resources. Method: This study was carried out in order to determine the most suitable explants and the best hormone combination to optimize micropropagation of linden in vitro. Explants were collected and cultured randomly from the best individual of linden from Gorgan Touskaestan forest. Findings: In light of results, from the eleven tested explants, stem containing terminal buds with scale were selected as the most suitable explants and IBA 0.1 mg/l was selected as the most appropriate medium hormonal combination in shoot regeneration. Discussion and Conclusion: Obtained data were analyzed with a completely randomized design by MSTATC software. It is hoped that the achieved results can be reproduced and be helpful in tissue culture research and also the molecular basis studies
