ارزیابی اثرات حفاظتی عصاره هیدروالکلی دانه عدسالملک (Securigera Securidaca L.) بر تغییرات پارامترهای بیوشیمیایی و شاخصهای کلیوی سرم در موشهای صحرایی نر تیمار شده با تامسولوسین

دانه عدس الملک سرشار از پلی فنل ها و فلاونوئیدها که اثرات بیولوژیک فراوانی دارند. هدف از انجام این مطالعه بررسی تاثیرات حفاظتی عصاره هیدروالکلی دانه گیاه عدس الملک بر تغییرات پارامترهای بیوشیمیایی سرم شامل گلوکز، پروفایل لیپیدی (کلسترول، تری گلیسرید، LDL و HDL)، شاخص های عملکردی کلیوی (کراتینین و BUN) و فعالیت آنزیمهای آنتی اکسیدانی (کاتالاز، سوپراکساید دیسموتاز و گلوتاتیون پراکسیداز) در موش های صحرایی نر تیمار شده با تامسولوسین بود. 72 موش صحرایی نر بالغ به 9 گروه هشت تایی گروه بندی شدند. گروه کنترل تیمار دارویی دریافت نکرد اما گروه شم 1 میلی لیتر حلال عصاره، گروه کنترل مثبت 4/0 میلی گرم/کیلوگرم تامسولوسین، گروههای کنترل منفی 1، 2 و 3 به ترتیب 200، 400 و 800 میلی گرم/کیلوگرم عصاره و گروههای تجربی 1، 2 و 3 ابتدا 4/0 میلی گرم/کیلوگرم تامسولوسین و سپس به ترتیب 200، 400 و 800 میلی گرم/کیلوگرم عصاره به مدت 28 روز دریافت نمودند. در انتها، نمونه های خونی جهت اندازه گیری پارامترهای بیوشیمیایی سرم جمع آوری شد. تامسولوسین باعث افزایش سطح گلوکز، کراتینین و BUN و کاهش سطح فعالیت آنزیمهای آنتی اکسیدانی در گروه کنترل مثبت شد (05/0p <) اما، تغییرات معناداری در سطح پروفایل لیپیدی ایجاد نکرد (05/0p >). با این حال، تیمار با دوزهای 400 و 800 میلی گرم/کیلوگرم عصاره در گروه های تجربی 2 و 3، موجب بهبود سطح گلوکز، کراتینین، BUN و فعالیت آنزیم های آنتی اکسیدانی در مقایسه با گروه کنترل مثبت گردید (05/0p <). عصاره هیدروالکلی دانه گیاه عدس الملک می تواند تغییرات پارامترهای بیوشیمیایی و شاخص های عملکردی کلیوی را در سرم موش های صحرایی نر تیمار شده با تامسولوسین بهبود دهد.

Securigera securidaca L. seeds are rich in polyphenols and flavonoids that have many biological effects. The aim of this study was to evaluate the protective effects of hydroalcoholic extract of Securigera securidaca seed on alterations in serum biochemical parameters including glucose, lipid profile (cholesterol, triglyceride, LDL and HDL), renal function indices (creatinine and BUN) and antioxidant enzymes activity (Catalase, superoxide dismutase, and glutathione peroxidase) in tamsulosin-treated male rats. Seventy-two adult male rats were grouped into 9 groups of 8. The control group did not receive drug treatment but the sham group received 1 ml of extract solvent, the positive control group received 0.4 mg/kg Tamsulosin, negative control groups 1, 2, and 3 received 200, 400, and 800 mg/kg extract, respectively and experimental groups 1, 2 and 3 first received 0.4 mg/kg Tamsulosin and then 200, 400 and 800 mg/kg of extract for 28 days, respectively. Finally, blood samples were collected to measure the serum biochemical parameters. Tamsulosin increased the levels of glucose, creatinine and BUN, and decreased the level of antioxidant enzymes activity in the control group (p < 0.05), but did not cause significant changes in the level of lipid profile (p > 0.05). However, treatment with doses of 400 and 800 mg/kg of extract in experimental groups 2 and 3, improved glucose, creatinine, BUN and antioxidant enzymes activity compared to the control group (p < 0.05). Hydroalcoholic extract of Securigera securidaca seeds can improve changes in biochemical parameters and renal function indices in the serum of tamsulosin-treated male rats.
