ارزیابی سطح بیان گیرنده CXCR4 پس از خاموش کردن توسط siRNA در سلولهای سرطانی MDA-MB-468

چکیده مقاله :
مطالعات بسیاری نشان دادند که siRNAها میتوانند سبب خاموشی ژنهای مرتبط با رشد و تهاجم سلولهای سرطانی گردد. در عین حال؛ اثر دقیق siRNAها بر بیان ژن CXCR4 هنوز به طور کامل به اثبات نرسیده است. براین اساس مطالعه حاضر به ارزیابی سطح بیان گیرنده CXCR4 پس از خاموش کردن توسط siRNA در سلولهای سرطانی متاستاتیک سینه پرداخته و برسی هایی صورت گرفته است. طی این تحقیق تجربی-آزمایشگاهی سلولهای سرطانی سینه رده MDA-MB-468 توسطsiRNA اختصاصی ژن snail1 ترانسفکت شدند. جهت سنتز cDNA ابتدا از محیط کشت سلولی، Total RNA استخراج گردید سطح بیان ژن گیرنده CXCR4 توسط qRT-PCR ارزیابی شد. جهت انجام qRT-PCR از PCR panels مخصوص miRNA استفاده شده جهت کنترل داخلی و کالیبراتور داخل پلیت از Unisp6 RNA استفاده شد. تکثیر سلولها به وسیله آزمایش تریپان بلو بررسی شد. دادهها با استفاده از آزمون تی-تست تجزیه و تحلیل شدند. نتایج نشان داد ناکداون ژن snail1 توسطsiRNA سبب کاهش معنادار بیان CXCR4 در سلولهای سرطانی سینه رده MDA-MB-468 شد (05/0 p <). طبق نتایج حاصل از این پژوهش، ترانسفکت سلولهای سرطان سینه توسط siRNA اختصاصی ژن snail1 می تواند سطح بیان CXCR4 را کاهش داده و در نتیجه سبب کاهش تکثیر و تهاجم سلولهای سرطان سینه و به مرور زمان باعث کنترل بیماری گردد.
چکیده انگلیسی:
Many studies have shown that siRNAs can silent the genes involved in growth and invasion of cancer cells. However, the exact effect of siRNAs is not entirely clear on the expression of CXCR4 gene in breast cancer cells. Therefore, the present study evaluated the expression of CXCR4 gene after silencing the siRNA in metastatic breast cancer cells. In this experimental-laboratory study, MDA-MB-468 breast cancer cells were transfected by the specific siRNA of the snail1 gene. The expression levels of CXCR4 gene were evaluated by qRT-PCR. Cell proliferation was assessed by trypan blue test. Data was analyzed using t-test. Snail1 knockdown by siRNA significantly reduced the expression level of CXCR4 in MDA-MB-468 breast cancer cells (P<0.05). The results of this study showed that the transfection of breast cancer cells by the specific siRNA of snail1 gene can reduce the expression level of CXCR4 and thus reduce the proliferation and invasion of breast cancer cells.
